Cinnabar having the efficacy of cooling pericardium and tranquilization was used widely in many Chinese patent medicines. During clinical application, cinnabar was used as dissolved mineral powder by improperly cooking, which resulted to large consumption and toxic side effects, thus affected the applications of cinnabar and cinnabar-containing combination drugs in medical field.Based on the above reasons, cinnabar was leaching treated by highly active thiobacillus ferrous oxide isolated in this topic, in order to dissolve the main element Hg and various trace elements. The anticonvulsant efficacy tests of the cinnabar leaching fluid were performed to detect whether the cinnabar leaching fluid had the traditional anticonvulsant effect. This could systematically analyze the functionary conformation, mechanism, formative principal and toxic mechanism of cinnabar in Chinese patent medicines, then supply substance basis and premise conditions for secondary development of cinnabar drugs.The results of experiments showed the optimum cinnabar leaching condition was: pHl.5-2.0, the content of ferric salt1%, ore concentration1%. The optimum shake flask time was25days, on this condition the Hg content of leaching fluid was2.8μg/ml.The cinnabar leaching fluid toxicity tests showed the drug maximum tolerance of leaching fluid was6.72ug, this dose was so little that the toxicity was minimal.The pharmacological experiment of cinnabar leaching fluid showed:the cinnabar leaching fluid could play better effect with little Hg content. The effect of cinnabar leaching fluid on convulsion latency induced by strychnine nitrate, pentylenetetrazol, benzoic acid, sodium caffeine was significantly higher than the traditional cinnabar suspension, the latency of convulsion was respectively(s):183±55、96±3、96±15, there was significant difference compared with the cinnabar suspension (AP<0.05).and the large dose group had the best effect during the three anticonvulsant models. In a word, the significantly anticonvulsant effect of cinnabar leaching fluid was observed, and the mortality induced by strychnine nitrate and sodium caffeine could be reduced.Children sedatives, an ancient recipe used for children convulsion, stopped to produce in2008due to serious side effects during use in recent years. In this study, cinnabar mineral drugs were replaced by cinnabar leaching fluid, a new type of children sedatives was developed, and pharmacodynamics experiment was implemented. The results showed, the efficacy of the new type of children sedatives was equivalent with positive drug (common children sedatives). The convulsion latency induced by strychnine nitrate, pentylenetetrazol, benzoic acid, sodium caffeine, the latency of convulsion was respectively(s):108.2±25、109.3±2、116±19, there was significant difference compared with the blank group,and the curative effects was similar with positive control group.the experiment showed the new children infantile convulsion powder could prolonged the latency of convulsion significantly, and the mortality of mice induced by strychnine nitrate and sodium caffeine was reduced, but the convulsion rate was100%,so the convulsion rate of three drugs could not be reduced. Against the convulsions caused by the sodium benzoate, caffeine, new children sedatives (containing cinnabar leaching fluid) to extend convulsion latency was significantly superior to traditional children sedatives. Therefore, the new type of children sedatives could play a good anticonvulsant effect with little toxicity.
