Based on the analysis of the existing production process of diosgenin from Dioscorea zingiberensC.H.Wrigh, research of high efficiency extraction methods were conducted to overcome its shortcomingssuch as high pollution and high energy consumption. The main content of this dissertation includes:1.The intensifying effect of ulatrasonic radiation on the extraction of dioscin from Dioscoreazingiberensis C.H.Wrigh was studied systematically using orthogonal design and single-factor experiments.The influence of major factors on the extraction of total saponins, such as ultrasonic frequency, extractingtime, ultrasonic power, extracting temperature, concentration of SDS, and the ratio of extracted solvent tomaterial, was examined. The optimized extracting conditions were found to be: ultrasonic frequency20KHz, extracting time5min, ultrasonic power100W, extracting temperature15℃, W(SDS)2%, the ratioof extracted solvent to material10:1(mL/g).2. The two aqueous phase system composed of HCl-surfactant-water and petroleum was used toperform in-situ hydrolysis and extraction of dioscin from Dioscorea zingiberensis C.H.Wrigh. The optimalconditions were found to be: extraction temperature80℃, Hydrochloric acid concentration2mol/L,extracting time3h, W(SDS)3%, the liquid-solid ratio16:1, petroleum ether90mL. The yield of diosgeninwas higher than that of traditional acid hydrolysis and methanol extraction methods, the extraction timewas also reduced significantly, and the operation was simplified.3. The effect of microwave and ultrasonic on extraction using two aqueous phase was studied. Bothmicrowave and ultrasonic were found to be capable of enhancing the extraction process. The effects of some single factors such as microwave time, ethanol and ammonium sul-fate concentration were alsoconducted.
