CCK-8assay was used to assesse the cell growth inhibition andcompare the differences between them for statistical significance. Fluorescent microscopewas used to observe morphological changes of apoptosis of SMMC-7721hepatoma cancercell after added lenalidomide or thalidomide. Flow cytometry was used to detect the effectof lenalidomide and thalidomide on SMMC-7721hepatoma cancer cell apoptosis.Enzyme-labeling instrument was used to measure the activity of Caspase-3. ELISA wasused to detect the expression of vascular endothelial growth factor(VEGF) of SMMC-7721hepatoma cancer cell after added thalidomide or lenalidomide.Results: After lenalidomide or thalidomide was added to SMMC-7721for48h,thephenomenon was detected by inverted microscope that the cells became less with thehigher concentration,while suspension cell became more in the supernatant.Boththalidomide and lenalidomide could inhibit the proliferation,induce the apoptosis ofSMMC-7721in a dose-dependent manner. What,s More, the effect of lenalidomide wasmore stronger than thalidomide with the same concentration. The mechanism of inducingapoptosis may associate with Caspase-3. The VEGF expression levels in SMMC-7721hepatoma cancer cells were reduced after the treatment of thalidomide or lenalidomide.Also the effect of lenalidomide was more stronger than thalidomide with the same concentration.Conclusions: Lenalidomide can inhibit the proliferation, induce apoptosis of theSMMC-7721cells and inhibit the expression of VEGF in vitro. The effect of lenalidomideis more potential than that of thalidomide. The mechanism of inducing apoptosis mayassociate with the activation of caspase-3.
