Objective:Our study examined the cell inhibition ratio and the expression levels of4E-BP1、 S6K1、 mTOR and VEGF in Hela cell,which was stimulated with different concentration Panax Notoginsenoside.We are expecting that this study will contribute to prove the machinery that Panax Notoginsenoside suppress cervical cancer,and may therefore provide a potential therapy for human cervical cancer.Methods:In order to find out the changes of cell inhibition ratio and the expression of luciferase reporter gene,p-S6Kl,p-4E-BPl,p-mTOR, S6K1,mTOR,4E-BP1,VEGF in Hela cells,We analyzed the data by MTT experiment and Western-Blot,luciferase reporter gene technology and ELISA.Results:(1)MTT experiment shows:the growth of Hela cells were significantly inhibited by Panax notoginsenoside(P<0.05).And dramatic difference was found in different dose and time (p<0.05).(2)The differences of LUC was statistically significant between experimental group and its controller by luciferase reporter gene technology(P<0.05).(3) S6K1,4E-BP1,mTOR and the level of their phosphorylation was detected by western-blot.After stimulated with different Panax Notoginsenoside,S6K1,4E-BP1,mTOR, p-S6Kl,p-4E-BP1and p-mTOR were distinctly decreased in the concentration of200Μμg/mL(p<0.05).(4)The OD value of VEGF in experimental group was lower than the control group, and the difference was obviously significant (p<0.05).Conclusion:The growth of Hela cells were significantly inhibited by Panax Notoginsenoside.we supposed that it happened through the mTOR signaling pathway.The efficiency of mRNA translation, the expression of VEGF,and the formation of translation initiation complex eIF4F were inhibited,which affect the composition of protein and prevent the development of uterine cervix cancer.It may therefore provide a potential therapy for human cervical cancer.
