ObjectiveBy establishing the transient common bile duct obstruction model and renal ischemia-reperfusion injury in mice, we the effect of transient common bile duct obstruction on liver and kidney function, as well as the expression of heat shock protein27and hemeoxygenase-1in liver and kidney. We explore the mechanisms of action induced by transient common bile duct obstruction on ischemia reperfusion injury, to provide experimental basis for the interaction between the liver and kidney. MethodsFirst, establish the model of the transient common bile duct obstruction.2days later, loosen the ligature. We measure the serum total bilirubin, alanine aminotransferase, creatinine at the indicated0d、1d、2d、3d、4d、5d and the expression of heat shock protein27and hemeoxygenase-1in liver and kidney at the indicated3d、4d、5d after common bile duct ligation. There are five mice at each time point. Establish renal ischemia reperfusion model with right nephrectomy and left kidney for ischemia25min and reperfusion24h. There are40mice divided into four groups. SHAM+NOIR group:just sham surgery is done without kidney ischemia/reperfusion,5days later, collect the specimens; SAHM+IR group:4days after sham surgery, kidney ischemia/reperfusion is done; CBDL+NOIR group: transient common bile duct obstruction model is done without kidney ischemia/reperfusion; CBDL+IR group:4days after transient common bile duct obstruction surgery, kidney ischemia/reperfusion is done. We measure serum creatinine and do paraffin-embeded renal specimens in each group. Do PAS staining of kidney specimens to analyze morphological changes of kidney tissue, MPO immunohistochemical staining to analyze neutrophil infiltration of the kidneys and TUNEL staining to analyze renal cell apoptosis in each group. Experimental data are presented as mean±standard deviation(x±s)and are analyzed by SPSS10.0statistic analysis software. Multiple sets of data are compared with single factor analysis of variance; and we make LSD-t test compare groups, witha=0.05level as a significant test.ResultsAfter the common bile duct ligation, the serum total bilirubin and alanine amino transferase increase rapidly and then decline rapidly after loosening the ligature; there is no change in serum creatinine. The expression of heat shock protein27but not hemeoxygenase-1in liver and kidney at the indicated3d、4d、5d after common bile duct ligation is upregulated. Compared with SHAM+IR group, CBDL+IR group has significantly lower serum creatinine (P<0.05), reduced renal tissue injury (P<0.05), decreased neutrophil infiltration(P<0.05) and less apoptotic cells(P<0.05). ConclusionsTransient common bile duct obstruction in mice impairs seriously the liver function, but the kidney function. Transient common bile duct obstruction upregulates the expression of heat shock protein27in liver and kidney. Transient common bile duct obstruction can attenuate kidney ischemia/reperfusion injury in mice, probably through promoting the expression of HSP27protein.
