Objective: The rat limb ischemia-reperfusion injury(ischemia-reperfusion injure,I/RI)animal models were established to observe the curative effect of Xuebijing on rat limbischemia-reperfusion injury while the reperfusion Xuebijing was injected in rat. To study theeffect of Xuebijing on limb ischemia-reperfusion injury and to lay the foundation for furtherdevelopment of the Xuebijing clinical was used to prevent and cure limb ischemia-reperfusioninjury.Methods:The experimental models of Lower limb ischemia/reperfusion injury wereestablished in 36 Wistar rats that were random divided into three groups,every group have 12Wistar rats. Every group adopted ventral median abdominal incision. The sham operation grouponly opening abdominal and detaching aorta abdominalis, don’t clamp aorta abdominalis. Theischemia-reperfusion group and XBJ group were used noinvasive arteryclamp to clamp aorta 2hours. As reperfusion normal saline and Xuebijing injection was infused into rats caudal veinrespectively. Then recoved bloodstream 4 hours. After 4 hours reperfusion,the samples of bloodwere taken from caudal vein respectively for measurement of MPO, LDH and NOS(TNOSã€iNOSã€cNOS)serum activity. After HE dyed, skeletal muscle and blood vessel wall structurechanges were observed by light microscope.Results:As compared with those in sham operation group(SOG), the serum activity ofMPO, LDH, TNOS, iNOS were rose in both ischemia-reperfusion group(I/R) and XBJ group(P<0.05).The serum activity of cNOS was reduced in I/R group(P<0.05). The serum activityof cNOS was reduced in XBJ group(P<0.05). As compared with those in I/R group, the serumactivity of MPO, LDH, iNOS were reduced in XBJ group(P<0.05). The serum activity of TNOSand cNOS activity was rose in XBJ group(P<0.05). After 4 hours reperfusion, in the I/R groupskeletal muscle cells appeared swell, intercellular space narrow down; And a number ofinflammatory cells infiltrated the intercellular. Muscle fiber oedema, arranged irregular and partof muscle fiber appeared fracture; Each layer of blood vessel wall arranged loose, cell oedema.VECs combine with endothelium under elastic membrane became loose; Part of the VECs shedto lumen, and inflammatory cells clinged. Compared with the I/R group, in the XBJ groupskeletal muscle cells swell alleviated, Muscle fiber oedema alleviated, arranged tidiness and partof muscle fiber appeared tortuosity; VECs arranged tightness, cell oedema alleviated. VECscombine with endothelium under elastic membrane tightness; Few of the VECs shed to lumen,and less inflammatory cells clinged. Conclusion:The Xuebijing injection has protective effect on ischemia-reperfusion injury oflower limbs in rat by stabilizing neutrophils, reducing the iNOS activity, and decreasing theiNOS-derived NO while rising the cNOS activity, and increasing the eNOS-derived NO. |
