| Objective(1) Using the Bethesda method to detected the positive rate of factor â…§ inhibitor in severe hemophilia A.(2) To investigate the potential association between factor â…§ inhibitor development and polymorphisms of human leukocyte antigen-DQAl(HLA-DQA1) gene in patients with hemophilia A.methods(1)All of the patients plasma samples were measured by Bethesda method.(2)The alleles polymorphisms in HLA-DQA1gene was analyzed in130patients with hemophilia A and90normal controls by using polymerase chain reaction-sequence specific primer(PCR-SSP).Results(1) In130cases of HA patients, Fâ…§ inhibitor was detected in6patients from totally88patients (6.82%).(2) In130cases of HA patients,10alleles gene frequencies was from3.85%-65.38%. In90cases of normal control group,10alleles gene frequencies was from3.33%-63.30%.(3) Two groups were defined, that one group included all of the HA patients, other group included severe patients, we used^test analysis to compared the occurrence rate of alleles gene in HLA-DQA1gene region of these two groups separately to the rate of HA patients who were detected inhibitors. The results respectively showed that the difference were insignificant (P>0.05).Conclusion(1) The positive rate of inhibitor in130cases of patients with HA was lower than those in literature reports. The causes of Fâ…§ inhibitor occurrence is needed to be further research.(2) The phenomenon of HLA-DQA1gene polymorphism is widespread in our country, but the alleles polymorphisms in HLA-DQA1gene associated with Fâ…§ inhibitor occurrence cannot be determined at present. The formation of inhibitor has multiple factors, genetics plays a major role in this mechanism. |
