Background:Pituitary adenoma is one of the common neuroendocrine tumors, accounting for1/10ofintracranial tumors. Pituitary tumor is a kind of benign tumors, but part of pituitary adenomawith malignant tumor biological characteristics, manifestations of invasive growth, to theintracranial around the sellar region and sphenoid sinus tissue invasion, intracranial vascularand nerve damage. Research confirms, pituitary tumor invasion is the main factor affectingpostoperative recurrence. Pituitary tumor recurrence rate is high up to50%~60%. Pituitarytumor recurrence great harm patients, treatment will be more difficult, to bring patients a lotof difficulties. The pituitary tumor invasiveness and recurrence problems should be a widerange of clinical heavy attention. The histopathologic examination of cell morphologycannot distinguish between noninvasive and invasive pituitary adenoma, then, at the proteinlevel on in-depth study of invasive pituitary tumor occurrence, development and recurrencemechanism, will be helpful to the improvement of clinical diagnosis, differential pituitarytumor, In order to make its recurrence trend forecast, and take the corresponding individualtreatment regimens to prevent or reduce the recurrence of pituitary tumor.Purpose:This paper through the detection PTTG, Ki-67and P16, P27protein expression ininvasive pituitary adenoma and non invasive pituitary tumor, and analysis correlation ofPTTG, Ki-67and P16, P27and pituitary adenoma recurrence, so as to explore the PTTG,Ki-67and P16, P27gene in invasive pituitary adenoma recurrence occurred and thesignificance.Methods:Pituitary tumor tissue specimens from2007January to2008December neurosurgeryGeneral Hospital of Daqing Oil Field hospitalized patients with pituitary tumor operation,patients with complete clinical data, treatment and follow-up results, a total of60patients,divided into two groups, in which invasive pituitary adenomas in32cases (including24cases relapsed), and non invasive pituitary adenomas in28cases (including8cases ofrecurrence). All cases with recurrence diagnosis used uniform standards. Immunohistochemistry (Streptavidin Perxidase,SP) were determined PTTG, Ki-67and P16,P27expression in two groups of patients with pituitary tumors in paraffin sections, andstatistical data, analysis of PTTG, Ki-67and P16, P27expression intensity in two cases ofdifferences, further analysis of their correlation with pituitary adenoma recurrence.Results:In32cases of invasive pituitary adenoma, the expression of PTTG was positive in26cases,18cases were strongly positive,8cases were weakly positive; The expression of Ki-67was positive in22cases;16cases were strongly positive,6cases showed weak positive; In28cases of invasive pituitary adenoma, the expression of PTTG was positive in9cases,2caseswere strongly positive,7cases weakly positive; Ki-67expression was positive in6cases,2cases were strongly positive,4cases showed weak positive; PTTG and Ki-67expression ininvasive pituitary adenoma was significantly higher than that of non invasive tumors;32casesof invasive pituitary adenoma, the expression of P16was positive in8cases,4cases werestrongly positive,4cases were weakly positive; The expression of P27positive was7cases,4cases were strongly positive,3cases showed weak positive; In28cases of invasive pituitaryadenoma,24cases were P16positive,17cases were strongly positive,7cases showed weakpositive; P27expression was positive in23cases,18cases were strongly positive,5casesshowed weak positive; P16and P27expression in invasive pituitary adenomas was lowerthan in non invasive tumors; and confirmed invasive pituitary adenoma in presence of PTTG,Ki-67gene high expression and P16, P27gene low expression. In the present experiment,the PTTG, Ki-67and P16, P27gene expression and pituitary adenoma recurrence relation toundertake an analysis, found that PTTG and Ki-67genes and pituitary adenoma recurrencewere positively correlated, P16gene and pituitary adenoma recurrence was negativelycorrelated.Conclusions:The expression of PTTG and Ki-67in invasive pituitary adenomas was higher than noninvasive tumors, PTTG and Ki-67was involved in pituitary tumor invasive growth process;P16and P27in invasive pituitary adenoma were lower than levels of non invasive tumors,P16and P27were also associated with pituitary tumor invasive growth; The expression ofPTTG and Ki-67genes and pituitary adenoma recurrence were positively correlated, Theexpression of P16gene and pituitary adenoma recurrence was negatively correlated withPTTG and Ki-67, showed increased expression facilitates pituitary tumor recurrence, P16expression decreased contribute to pituitary adenoma recurrence. Through the study of this subject, confirmed cancer gene PTTG, Ki-67and P16tumor suppressor gene P27in tumorinvasiveness, vertical formation and recurrence process, and can be used as a heraldingpituitary tumor with invasiveness and recurrence after the possibility of biological indicators,to a certain extent, can guide the pituitary tumor individualized comprehensive treatmentscheme, in order to improve the clinical efficacy of pituitary tumor.
