Stat3, a member of STATs(signal transducers and activators of transcription,STATs)family have been shown to play a key role in promoting proliferation, differentiation,anti-apoptosis or cell cycle progression。P-glycoprotein(P-gp) encoded by MDR1gene is one of the most important reasons forMultidrug resistance(MDR).The human P-gp(Mdr-1) on cell membrane as efflux pump outantitumor agents from the cytoplasm reducing intracellular drug concentrations in the cells.RNA interference (RNAi) is the process of sequence-specific posttranscriptional genesilencing triggered by double-stranded RNAs(dsRNAs) homologous to the silenced gene。We Constructed recombinant plasmids of co-expression of MDR1-of SiRNA andSTAT3-of SiRNA,lipofectamine2000(Invitrogen) was used as the transfection to Transfectthe recombinant plasmids into the Human lung adenocarcinoma cel(lA549),RT-PCR、MTTand Western blot analysis were used to detect the expression of the MDR1and Stat3proteinand mRNA levels,the results showed recombinant plasmids could significantly inhibited themRNA and protein level of STAT3and MDR1expression. As we all know MDR1is drugresistance gene, our experiments will be provide a theoretical basis and preparatory work forcancer gene therapy with chemotherapy drugs。...
