| Part IEeffects of simvastatin on the expression of toll-like receptor4ã€nuclear factor-κB and interleukin-1after acute spinal cord injury in ratsObjective:To observe the influence of simvastatin administration on expression of toll-like receptor4(TLR4)ã€nuclear factor-kappa B(NF-κB) and interleukin-1β (IL-1β) following acute spinal cord injury so as to explore the protection mechanisms of simvastatin in the spinal cord injury.Materials and methods:30SD rats were randomly divided into3groups:Control group(sham),spinal cord injury group (SCI) and treatment group (Simvastatin), n=10.Treatment group received simvastatin (37.5mg/kg) p.o. at1and6h after surgery..All the rats were sacrificed at24h after trauma,the expression of TLR4protein were assessed by western-blot,EMSA assay the DNA-binding activity of NF-κB and ELISA assay the expression of IL-1β.Results:The levels of TLRã€NF-κB and IL-1β in spinal cord tissue were significantly inhibited in the treatment group than the injury group (P<0.05) Conclusions:Simvastatin administration can down-regulate the expression of TLR4ã€NF-κB and IL-1β to inhibit the inflammatory response following SCI, and play a protective role on the secondary spinal cord injury. |
