Objective:SOCS3was short for suppressor of cytokine signaling3,it played an important role in regulating organs injuries of diseases,and it could inhibite cytokines from releasing by feedback regulation of JAK/STAT signal path,however,there was no report about the role of SOCS3in inhibiting inflammation reaction in severe acute pancreatitis.We aimed to determine the expression of SOCS3in pancreas and lung tissue of rat with severe acute pancreatitis and detemine changes of cytokines in serum to investigate the mechanism of action of suppressor of cytokine signalling,3(SOCS3) in experimental severe acute pancreatitis(SAP).Methods:The rat model of SAP was reproduced by retrograde injection of4%sodium taurocholate into the biliopancreatitc duct.Thrity-two male SD rats were randomly assigned into4groups (8each):normal control group (NC),SAP6h,12h, and18h groups. The level of serum amylase (AMY) was measured dynamically.The pathological changes of pancreas and lung were observed under the light microscope, and the wet/dry weight ratio of lungs were calculated.The concentration of IL-6、IL-18were determined by ELISA. The localization and expression of SOCS3protein in pancreas and lungs were determined by immunhistochemical staining and Western bloting. Result:Compared with NC group,serum level of AMY icreased significantly in SAP group (P<0.05), the pancreas and lung injuries under the light microscope were gradually aggravated with disease progression, and the wet/dry weight ratio of lungs increased significantly compared with NC group (P<0.05).The serum concentration of IL6、IL-18in the SAP groups increased significantly compared with NC group (P<0.05). The concentration of SOCS3protein in NC group is little,The concentration of SOCS3protein in SAP groups increased significantly compared with that in NC groups (P<0.05).The change of SOCS3protein concentration is in concidence with the severity of pancreas and lung tissue as well as the serm concentration of IL-6、IL-18.Conclusions:The inflammatory action induced by SAP can induce the expression of SOCS3in pancreas and lung tissue, of which increases accompany to the severity of lung injury and inflammatory action. It is suggested that there is maybe certain connection with negative feedback role of SOCS3in inflammatory action mediated by JAK/STAT signaling pathway.
