Objective Observe the expresssion of BMI-1and P14ARF in NSCLC andinvestigate its mechanism and clinicopathologic significance, and analyze therelationship between BMI-1gene expression and P14ARF. We aim to investigate theexpresssion ot BMI-1, P14ARF in non-small cell lung cancer and to explore itscorrelation with the clinical pathological characteristics and its influence on patients’prognosis.Methods Determine the expression of BMI-1and P14ARF in NSCLC by Envisionimmunohistochemistry method in tissue microarry prepared from130NSCLC patients.Statistical analysis was performed to explore the BMI-1and P14ARF correlation withthe clinical pathological characteristics. Categorical data were calculated aspercentages and compared using the R×C test. The correlation between BMI-1andP14ARF was analyzed by Bivariate Correlation. Survival curves were plotted with theKaplan-Meier formula from the date of surgery as the starting point, the log-rank testwas performed for the curves.Results1)The immunohistochemical study showed that the positive expression ofBMI-1and P14ARF protein in130NSCLC patients were72.3%and47.7%;2)Theexpression of BMI-1was determined to associated with the TNM stage, lymph nodemetastasis, histology condition. However, its relationship with sex, age, tumor size,surgery procedure were not established (p>0.05). Also, the expression of P14ARF wasdetermined not to be associated with the patient’s sex, age, lymph node metastasis,histology,tumor size and surgery procedure(p>0.05). However, its relationship werehighly correlated with TNM stage.3)From our study we found that the the expressionof P14ARF was significantly higher in Stage I when compared with Stage II and III.4)BMI-1expression and P14ARF expression were negatively correlated(r=-0.510,p<0.001).5)Furthermore, it was shown that the survival rate of patients with negativeBMI-1expression was significantly higher than that of those with positive BMI-1 expression(p=0.000). In addition, the survival rate of patients with positive P14ARFexpression was significantly higher than that of those with negative P14ARFexpression(p=0.001).Conclusions We considered that there was high expression of BMI-1in NSCLC,which played an important role in lung cancer development and invasion via P14ARFgene expression protein and combined with the underexpression of P14ARF wasclosely related to the patients’ clinical pathological characteristics.
