| Objective Endometrial cancer is one of the most common female cancers, and it seriously endangers the health of women. So Study on the therapeutic approaches and treatment mechanism of endometrial cancer is of great significance. Currently, chemotherapy is an important adjunctive treatment of endometrial cancer, carboplatin is one of the effective chemotherapy drugs. In this study, we investigate the effect of carboplatin on the biological behaviour of endometrial cancer cell line, and to explore the possible mechanism of its effects. It may provide some information to comprehend the mechanism of carboplatin treatment.Methods1. Ishikawa cells were treated with different concentrations of carboplatin.2. Western blot analysis of bcl-2and bax expression was performed in Ishikawa cells line.3. Flow cytometry(FCM)was performed to analyze the cell apoptosis and cell cycle.4.Transwell invasion experiments were performed to evaluate the invasive activity. Results1. Treatment of carboplatin suppressed the Ishikawa cells growth.2. The expression of bcl-2protein downexpressed and the expression of bax protein increased after treatment of carboplatin.3. Treatment of carboplatin increase the cell apoptosis and cells cycle arrest in G2phase.4. Treatment of carboplatin suppressed the Ishikawa cells invasion.Conclusion This study indicated that Carboplatin can significantly affect endometrial carcinoma Ishikawa cell line biological behavior, This effects may be accomplished by down-regulating the ratio of bcl-2/bax. |
