The Study Of The Nourishing Kidney Methods On Inducing Bone Marrow Mesenchymal Stem Cells Into Chondrocytes And Observation Of Clinical Outcome For Treating Knee Osteoarthritis

ObjectiveTo investigate the effect and mechanism of icariin on chondrocyte proliferation and differentiation. And To observe the outcome of the norishing kidney methods for treating the early and middle stages of osteoarthritis.MethodsThe third generation human bone marrow mesenchymal stem cells (hBMSCs) were cultured and Collected in vitro, dividing into five groups:the Icariin groups, including low-dose, medium-dose, high-dose (concentration of20μg/L,40μg/L,60μg/L respectively); control group, adding the induction medium (TGF-β10μg/L, Dex0.1μmol/L, VitC50mg/L);and the blank group. HBMSCs proliferation and differentiation was detected by MTT method after1,3,5,7,9days. Identification of chondrocyte was checked by toluidine blue staining and Immunohistochemical method after culturing14-21days. Clinical aspects:40cases, meeted the standards of the early and mid-knee osteoarthritis diagnosis patients, were recruited for study. Self-control study was conducted.All the patients received Chinese patent medicines Xianling Gubao Capsule (the main ingredient was Icariin),3pieces one time, twice each day for6weeks. WOMAC index and traditional Chinese medicine syndrome wered compared before and after treatment. Using SPSS17.0for statistical analysis.ResultExperimental result:OD value in high-dose group (60μg/L) were highest, low-dose group lowest;OD value of third, fifth, seventh, ninth day of high-dose group was significant difference(P<0.05,), compared the blank group. OD value of third, fifth, seventh, ninth day in Medium-dose group and low-dose group were also high than the blank group, but P>0.05, not significant difference.Clinical result:After treatment, Knee pain and function and WOMAC index were improved significantly, and there were significant difference (P<0.05).3cases (8.0%) were significantly effectived,33cases (82.5%) were effectived, and4cases (9.5%)were invalid, The total effective rate were90.5%.Conclusion1. Icariin significantly promotes hBMSCs proliferative and reduce hBMSCs into chondrocytes.2. The higher the concentration of icariin,, the better effect of inducing into chondrocytes between the concentration of20μg/L to20μg/L.3. Xianling Gubao Capsule can treat the early and mid-treatment of osteoarthritis of the knee effectively, And reduce pain and delay the progress of osteoarthritis effectively.
