Objective:To observe tetrandrine (Tet) on the preventive and therapeutic effects of acute radiation radiation-induced lung injury in rats to explore its feasibility as a preventive measure for acute radiation-induced lung injury, and discuss its mechanism.Methods:48cases of healthy SD rats were randomly divided into four groups: group A (Tet and radiotherapy group, injection of Tet30mg/kg-1.d-1), group B (radiotherapy group, NS2ml/kg-1.d-1), group C(dexamethasone and radiotherapy group, injection of dexamethasone lmg/kg-1.d-1), group D(control group, NS2ml/kg-1.d-1).7days before irradiation start injection, until14days after exposure to an end.7days after injection, rats in group A,group B and group C were irradiated simultaneously, a dose of15Gy to the whole thorax was delivered in a single fraction at the posterior field using a linear accelerator of Varian. Six rats randomly selected to kill from Group A,group B, group C and group D each time at the15d,30d after radiation. After executing the rats, HE and Masson staining to observe pathological changes of the lung tissue; ELISA detection of serum IL-la, IL-6and TNF-a and TGF-β1level; Immunohistochemical(Envision) detection of lung tissue IL-la, IL-6TNF-a and TGF-β1protein expression changes.Results:1. Histologieal change:15days after exposure,the alveolar walls mild thickening, pulmonary intersitital mild edema, alveolar walls visible a little blue collagen fibers in group A; The alveolar wall thickening, interstitial pulmonary edema, inflammatory cell, alveolar walls and lung interval see collagen deposition in group B:The alveolar walls mild thickening, pulmonary intersitital mild edema, alveolar walls visible a little collagen fibers in group C.30days after exposure, group A of alveolar walls, thickening between pulmonary edema, collagen deposition increased, lung between group B thickening of wide obvious, pulmonary edema serious, visible inflammatory cell and foam cells and there are lots of collagen fiber calm. Group C alveolar wall thickening, between pulmonary edema, increased collagen deposition. group A, C compared with group B the same period, the alveolar reduce inflammation, relieve edema, cellulose sample exudate reduce, collagen deposition decreases.2. The level of cytokines IL-la, IL-6, TNF-a and TGF-β1:2.1IL-la, IL-6, TNF-a, TGF-β1concentrations in serum:15days after exposure, group A, group B and group C the index are higher level than group D, and group B is obvious increased. Group B compared with group D have statistical significance (P<0.05). Group A of expression level minimum, and Group A compared with group B have statistically significant (P<0.05).30days after exposure, group A of expression level drops, group B and group C increase, group A, C compared with group B have statistically significant (P<0.05). Group B cell factor expression level is the highest, and compared with group D compared have statistically significant (P<0.05).2.2IL-1a,IL-6,TNF-a,TGF-β1protein expression of lung tissue:15days after exposure, group A, group C of lung tissue light yellow particles appear, a weak positive expression. Group B in the cytoplasm see tan particles of positive expressive objects, expressing strong. Group D express or weak don’t express;30days after exposure, group A, group B and group C express increase apparently. But group A and group C expression which is weak group B. Group D express or weak don’t express.Conclusions:1. The use of the Tet can reduce the radiation-induced lung injury in rats with acute inflammatory response.2. The Tet through inhibition of inflammatory cytokines IL-la, IL-6, TNF-a expression, thus inhibiting irradiation-induced inflammatory response. 3. The Tet can inhibition of TGF-β1expression has to reduce the radiation-induced lung fibrosis...
