| Background and objective: Tumor growth and metastasis rely on angiogenesis,blocking angiogenesis is an effective strategy to inhibit tumor growth. Angiogenicfactor vascular endothelial growth factor (VEGF) and its receptor vascular endothelialgrowth factor receptor2(VEGFR2) seem to play key roles in tumor-inducedangiogenesis. Endostatin specifically inhibits vascular endothelial cell proliferationand promotes apoptosis. None previous studies have researched the relationship of thethree, and their role as prognostic factors in NSCLC are inconclusive. In order toreveal this, we collected paraffin-embedded histological material from311NSCLCpatients for immunohistochemical studies. The sections were incubated with primaryantibodies to VEGFã€VEGFR2and ES. Expression of the immunohistochemicalstaining was assessed semi-quantitatively. Kaplan–Meier method and Coxproportional hazard model were used to evaluate the significance of VEGF andVEGFR2and ES expression for prognosis.Results: Immunohistochemistry showed that the expression of VEGF was foundto be35.7﹪, mainly distributed in cytoplasm of cancer cells; The expression of VEGF protein was closely related to the gender(P=0.003)ã€Histological type(p=0.003)ã€differentiation(P=0.05)ã€Tobacco use(P=0.003), but there was nocorrelation to ageã€tumor size and clinical stage. VEGFR2staining of the cytoplasmof malignant cells accounted for43.1%, Staining mainly located in the cytoplasm;The expression of VEGF protein was closely related to the gender(P=0.034)ã€Histological type(P=0.005)ã€Tobacco use(P=0.044), but no correlation with ageã€differentiationã€tumor size and clinical stage. ES mainly stained in cytoplasm of themalignant cells, accounted for50.2%, there was no correlation to genderã€ageã€differentiationã€tumor sizeã€clinical stage and tobacco use. ES was negative correlationto malignant lymph node metastasis(P=0.003);VEGF, VEGFR2and ES were positivecorrelation(P=0.000); survival time of patients with high ES expression were longerthan low expression ones, although no statistical significance(P=0.132).Conclusion: The expression of ES was significantly associated with NSCLClymph node metastasis, patients with low ES expression were inclined to lymph nodemetastasis; High ES expression probably defined patients with a favorable outcome;Expression of VEGF, VEGFR2and ES were positively correlation, supportedNSCLC patients benefited when combined using drugs of VEGF inhibitor and ES;VEGF and VEGFR2also predicted some clinical characters. |
