Objective:The purpose of this study was to research expression ofNFBD1in Nasopharyngeal Carcinoma,and the distribution of NFBD1protein in the Nasopharyngeal Carcinoma (NPC). To investigating theeffect of siRNA-mediated NFBD1depletion on chemosensitivity innasopharyngeal carcinoma cells,then the change of Chemosensitivity inthe NPC cells.Method:To collect specimens of nasopharyngeal carcinoma andnormol pharynx nasalis tissue at hospitals, the expression levels ofNFBD1in nasopharyngeal carcinoma and normol pharynx nasalis tissuespecimens were shown by RT-PCR and immunohistochemistry. theImmunohistochemical methods could also show the distribution of NFBD1protein in NPC. Using siRNA transfection to inhibit expression of NFBD1in Nasopharyngeal Carcinoma cell lines HNE-1(poorly differentiatedsquamous-cell carcinoma)and CNE-1(goodly differentiated squamous-cellcarcinoma), to detect expression levels of mRNA and protein of NFBD1by Real Time RT-PCR and Western-blot methods,the result of expressionlevels could show the effect of siRNA transfection, the cells were dividedinto w/o group, control siRNA group and NFBD1group. according to thegroups the cells were treated with diamminedichloroplatinum (DDP),survival rate of NPC cells were detection by MTT method.Result:The disparity of expression levels of NFBD1between nomalpharynx nasalis tissue and NPC was conspicuously, higher expression inNPC(P<0.05). the distribution of NFBD1was detection in the cytolymphand cell nucleus in NPC. NFBD1siRNA significantly inhibited theexpression of NFBD1, and compared with blank group, survival rate in theNFBD1siRNA group was greatly degraded after treatment withDDP(P<0.05).Conclusion:The NFBD1in the NPC is high expression.Thesynergistical effect of treatment with chemotherapeutic agents and NFBD1siRNA was apparently helpful to increase chemosensitivity innasopharyngeal carcinoma cells.
