Objective: To compare the expression level of regulatory T cells(Treg)in peripheral blood of patients with immune thrombocytopenia(ITP)before and after treatment,and to analyze the significance of Treg in thepathogenesis and treatment of ITP.Methods: According to the platelet count(Plt) in peripheral blood,34ITP patients were divided into three groups: mild group、moderate groupand severe group before treatment.28patients in the moderate group andsevere group received the standard regimen of glucocorticoid therapy andwere divided into responsive group and non-responsive grouppost-treatment. The expressions of CD4+CD25+Treg in peripheral blood ofITP patients in all groups and20health people (control) were accounted forthe percentage by flow cytometry(FCM)and analyzed by statistical.Results: The percentages of CD4+CD25+Treg in34ITP patientsincluding the mild group[(3.450.40)%]、the moderate group [(2.510.29)%]and the severe group[(1.340.78)%]and in5patientswith non-response post-treatment[(0.920.48)%]were obviouslydecreased than that in the controls(P<0.05), while the levels compared inevery two groups before treatment were significant difference (P<0.05).Meanwhile,before treatment,the level of CD4+CD25+Treg in responsivegroup was significantly higher than that in non-responsive group[(0.860.34)%](P<0.05).After treatment, the responsive casesin ITP patients were23and the non-responsive cases were5.CD4+CD25+Treg level in responsive group [(6.331.85)%]increasedcompared with that of the ITP patients before treatments[(2.010.80)%](P<0.05),but still lower than health controls(P<0.05). After treatment, theratios of CD4+CD25+Treg in non-responsive group did not obviouslychanged (P>0.05).Conclusion: The percentages of CD4+CD25+Treg in peripheral bloodof ITP patients is significantly reduced.While the account is decreased, thefunction is weakened,that may be related to the disorders of bothcell-mediated immunity and humoral-mediated immunity and play animportant role in the pathogenesis of ITP. Detection of CD4+CD25+Treg inITP patients may play a guiding significance in the study of pathogenesisand therapeutics.
