The change of cell cycle after using thiostrepton were analyzed by flow cytometry; Migration and invasion determinations were used to examine the effect of thiostrepton on the invasion and metastasis of Hep-2cell line.(2) RT-PCR was used to investigate the subtype of Foxm1. Immunocytochemistry, RT-PCR and Western blot was used to investigate the Foxml expression in Hep-2cell line by thiostrepton. Results The proliferation inhibition rate of thiostrepton on the Hep-2cell line after48h in each concentration group (1μM,2μMã€4μMã€8μMã€12μMã€16μM) are (5.27%ã€9.43%ã€20.25%ã€41.61%ã€67.96%ã€96.95%); Hep-2cell line were kept at stage G1/G0dose dependently in2μM and3μM group (P<0.05); The subtype is Foxm1C in Hep-2cell line. Foxml protein highly expressed in Hep-2cell line (P<0.05) were supressed successfully with thiostrepton in48h (P<0.05).Conclusion Thiostrepton inhibits the proliferationand, invasion, metastasis and FoxMl expression in Hep-2cell line is a dose-dependent manner. | | Keywords/Search Tags: | Thiostrepton, FoxM1, Hep-2, proliferation, invasion, metastasis | PDF Full Text Request | Related items |
