Objective (1) To explore the effect of thiostrepton on proliferation,invasion and metastasis in Hep-2cell line;(2) To explore the effect of Foxml in its way..Methods (1) MTT reduction assay was used to investigate the proliferation inhibition of thiostrepton on Hep-2cell line. The change of cell cycle after using thiostrepton were analyzed by flow cytometry; Migration and invasion determinations were used to examine the effect of thiostrepton on the invasion and metastasis of Hep-2cell line.(2) RT-PCR was used to investigate the subtype of Foxm1. Immunocytochemistry, RT-PCR and Western blot was used to investigate the Foxml expression in Hep-2cell line by thiostrepton.Results The proliferation inhibition rate of thiostrepton on the Hep-2cell line after48h in each concentration group (1μM,2μM、4μM、8μM、12μM、16μM) are (5.27%、9.43%、20.25%、41.61%、67.96%、96.95%); Hep-2cell line were kept at stage G1/G0dose dependently in2μM and3μM group (P<0.05); The subtype is Foxm1C in Hep-2cell line. Foxml protein highly expressed in Hep-2cell line (P<0.05) were supressed successfully with thiostrepton in48h (P<0.05).Conclusion Thiostrepton inhibits the proliferationand, invasion, metastasis and FoxMl expression in Hep-2cell line is a dose-dependent manner.
