| Objective:Use different doses of aspirin(ASA) intervention (Bone marrow stromal cells BMSCs)transplantation in rats with cerebral ischemia reperfusion injury, observation of serum TNF-a concentration, neurological function score, the expression of Caspase-3, to investigatethe effect of aspirin on BMSCs transplantation in rats with cerebral ischemia reperfusioninjury and the effect of mechanism.Methods:Made rat Middle cerebral artery occlusion (MCAO)models by the method of Suture(n=48).models were divided randomly as: Model natural recovery group (group A), BMSCs transplantationgroup(group B),Aspirin intervention group(group C),Aspirin intervention BMSCs transplanta-tion group(group D),groupC〠D was divided into three subgroups according to the dose ofaspirin,(Dose of aspirin:C1ã€D1subgroup3.75mg/kg/dã€C2ã€D2subgroup7.5mg/kg/dã€C3ã€D3subgroup22.5mg/kg/d),each subgroups in group Cã€D were Given different doses of aspirinintegra-ted into distilled water daily by gavaging in120hours after Reperfusion;group; A wasModel natural recovery group;groupBã€D was done BMSCs Transplantation, after Ischemia for2hours Reperfusion of146hours,2x106BMSCs was transferred into each SD rats via the tailvein. The fourteenth day we evaluated neurobehavioral score observed the cell morphology by HEstai-ning, determinated expressions of caspase-3.Results:1.The scores of neurologic function(unit:points):group A3.76±0.42,group B2.04±0.28,groupC:C1subgroup3.91±0.35〠C2subgroup3.83±0.62〠C3subgroup3.92±0.60,group D: D1subgroup1.69±0.16ã€D2subgroup1.17±0.11ã€D3subgroup2.25±0.17.group Ashowed a statistical difference compared with group B and each subgroup in group D (P<0.05),Group A was higher than group B and each subgroup in group D; group Ashowed no statistical difference compared with each subgroup in group C(P>0.05); groupB showed a statistical difference compared with each subgroup in group D(P<0.05),GroupB was higher than each subgroup in group D;subgroups in group D showed a statisticaldifference compared with each other(P<0.05),D3subgroup was highest,D2subgroup waslowest.2. Determination of Serum TNF-α by ELISA kits(unit: ng/ml):The scores of neurol-ogic function: group A7.69±0.54,group B6.27±0.42, groupC:C1subgroup5.59±0.20ã€C2subgroup5.59±0.20〠C3subgroup1.52±0.36, group D: D1subgroup4.76±0.52ã€D2subgroup2.47±0.36ã€D3subgroup0.75±0.18. group A showed a statistical differencecompared with group Bã€each subgroup in group C and each subgroup in group D(P<0.05),Group A was higher than group Bã€C and D;group B showed a statisticaldifference compared with each subgroup in group D(P<0.05),Group B was higher thangroup D; subgroups in group C showed a statistical difference compared with eachsubgroup in group D according to dose(P<0.05),Group C was higher than groupD;subgroups in group C showed a statistical difference compared with each other(P<0.05),C1subgroup was highest,C3subgroup was lowest; subgroups in group D showed astatistical difference compared with each other(P<0.05),D1subgroup was highest,D3subgroup was lowest.3. Brain histopathological slices stained with HE showed: Group A: nerve tissue oflarge areas of necrosis, surrounded by remnants of the organizational structure isincomplete, and edema of nerve cells, a large number of vacuolization organization edema,nerve cells increase in size, vacuolar degeneration, the cell gap increases; group B: in ratbrain tissuestructure is significantly improved, compared to group A necrotic area downinfarction around the remnants of the organization more; C: roughly the same rat braintissue necrosis and damage to the A; group D: cell damage significantly reduced comparedwith BMSCs transplantation group, infarctnarrow range, the cell is more complete, slightinterstitial edema, and D2sub-group of nerve cells is the most complete, D1, the Asiangroup, followed by D3subgroup is somewhat less in the D group. 4. Immunohistochemical staining was used to detect the expression of caspase-3(unit:cells): group A396.70±59.68, group B292.35±31.62,groupC:C1subgroup392.02±50.12ã€C2subgroup382.59±31.06ã€C3subgroup379.66±25.33,groupD:D1subgroup188.40±32.29ã€D2subgroup120.43±13.04ã€D3subgroup251.68±13.30. group Ashowed a statistical difference compared with group B and each subgroup in group D(P<0.05),Group A was higher than group B and each subgroup in group D; group Ashowed no statistical difference compared with each subgroup in group C(P>0.05); groupB showed a statistical difference compared with each subgroup in group D(P<0.05),GroupB was higher than each subgroup in group D;subgroups in group D showed a statisticaldifference compared with each other(P<0.05),D3subgroup was highest,D2subgroup waslowest.Conclusions:Aspirin intervention can promote BMSCs transplantation in rats with cerebralischemia reperfusion injury in a range of doses,may be associated with aspirin can reduceserum TNF-α levels of concentration of rats with MCAO reperfusion and increase BMSCslower damaged nerve of caspase-3expression. And promotion was dose related. |
