Objective To explore the fluctuation and its significance of dendritc cells includingplasmacytoid dendrtic cells (PDC) and myeloid dendrtic cells (MDC), cytokinesincluding IFN-,IL-6,TNF-in peripheral blood in patients with systemic lupuserythematosus (SLE).Methods Thirty-eight SLE patients,15PSS patients,15normal individuals wereenrolled in the study. Current classified criterias were fulfilled for all patients. All theclinic and laboratory data were collected. The propotions of PDC, MDC in peripheralblood were measured by flowcytometer. The concentration of IFN-, IL-6, TNF-wasdetected using ELISA. All the data were analysed by SPSS13.0.Results①T he level of PDC, MDC in peripheral blood of SLEpatient decreasedsignificantly comparing with normal controls((0.202±0.210%vs0.484±0.141%,P<0.05;2.004±1.139%vs4.313±1.047%, P<0.05). PDC, MDC levels from PSSpatients was lower than that in control group(0.228±0.277%vs0.484±0.141%,P<0.05;1.683±1.252%vs4.313±1.047%, P<0.05). But no evident difference in PDC,MDC wasfound between SLE patients and PSS patients(p>0.05).②It was also founded that thepercentage of peripheral blood PDC in SLE patients with stable disease wassignificantly higher than patient with active disease(0.05%vs0.20%, z=-2.722,p=0.011). SLE patients with proteinuria had lower level of PDC comparing withpatients without proteinuria(0.05%vs0.21%, z=-2.565, p=0.012). In addition,antids-DNA negative patients had higher percentage of PDC than anti ds-DNA positive patients(0.04%vs0.20%, z=-2.543, p=0.011). The levels of MDC in SLE patients withactive disease, patients with proteinuria, anti ds-DNA positive patients had nosignificant difference comparing with patients with stable disease, patients withoutproteinuria, anti ds-DNA negative patients(1.706±0.786%vs2.220±1.315%, p>0.05;1.795±1.067%vs2.304±1.203%, p>0.05;1.875±1.072%vs2.088±1.197%, p>0.05).③The ratio of PDC were correlated significantly with ESR, C3andSLEDAI,(r=-0.453, p=0.008; r=0.451,p=0.001; r=-0.363,p=0.031), but had nocorrelation with IgG、ANA titer、C4(p>0.05). The ratio of MDC were not correlatedsignificantly with ESR、C3、C4、SLEDAI、IgG、ANA titer(p>0.05).④We stillfound SLE patients secreted higher level of IFN-, TNF-,IL-6than controls(70.903±35.443μg/l vs39.056±14.169μg/l, t=3.138, p=0.003;71.010±29.392μg/l vs52.431±20.021μg/l, t=-2.066, P=0.046;37.129μg/l vs27.750μg/l, z=-2.743, p=0.006).The ratio of PDC,MDC had no correlation with levels of IFN-, TNF-,IL-6(p>0.05).⑤PSS patients also secreted higher level of IFN-than controls(99.452±55.056μg/l vs39.056±14.169μg/l, t=3.830, p=0.001).but there are no significant differentce ofTNF-, IL-6between PSS group and control(97.579±51.830μg/l vs52.431±20.021μg/l,p>0.05;58.474±54.272μg/l vs29.106±9.636μg/l, p>0.05). The ratio of PDC,MDChad no correlation.with levels of IFN-, TNF-,IL-6(p>0.05).Conclusion The ratios of PDC and MDC are decreased in PB of SLE and PSS. PDCdecrease significantly in disease active SLE. The decrease odf PDC were correlatedwith the activity of SLE and kidney damage. DC may be the cause of autoimmunediease. Level of IFN-, TNF-,IL-6in SLE patients were significantly increased,butno correlation with the PDC and MDC levels.
