Objective:To study the expression of Galectin-8and MMP-9in airways and lungs of mice with asthma and the effect of budesonide on the expression.Methods:Establish a mouse asthma model by sensitization and challenge with ovalbumin(OVA),thirty-six BALB/c SPF female mice were randomized into control group (group A),asthma group(group B) and budesonide intervention group(group C).In this experiment,the mice of group B and group C was sensitized by intraperitoneal injection of OVA and AI(OH)3on days1,8,15,and challenged on day22by aerosolised OVA for30min/day on three days/week for up to six weeks.The mice of group A were sensitized and challenged with saline. The mouse of group C were treated with budesonide (1mg, aerosol inhalation)1hours before challenged.Lung tissues were collected from mice in24hours after the last challenged. The pathological changes of the lung tissues were observed. The expression of Galectin-8and MMP-9protein were detected by immunohistochemistry(IHC).Galectin-8and MMP-9mRNA levels in lung tissues were determined using RT-PCR.Results:1.Pulmonary histopathology:In group B,light microscopy of HE-stained airways and lungs tissues of mice were showed airway epithelial damage and smooth muscle thickening.There was also evidence of inflammatory cells infiltrating around the airway and vessel. Masson stained lungs of group B showed lots of collagen deposition around the airway and vessel. Compared with group B,The pathological alterations in group C significantly alleviated.2.The results of Galectin-8Immunohistochemistry:Compared with group A (0.10%±0.05%), the protein expression of Galectin-8in group B (1.10%±0.39%) and group C(0.45%±0.15%) was significantly increased(P<0.05). Compared with group B, group C were significantly decreased(P<0.05).3.The results of MMP-9Immunohistochemistry:Compared with group A (9.91±1.50), the protein expression of MMP-9in group B(38.36±6.34) and group C (20.0±2.90) was significantly increased(P <0.05). Compared with group B, group C were significantly decreased(P <0.05).4.The results of Galectin-8RT-PCR:Compared with group A (0.47±0.03),the expression of Galectin-8mRNA in group B (0.79±0.61) and group C(0.72±0.29) were significantly increased(P<0.05). Compared with group B, the expression of group C were significantly decreased(P <0.05).5. The results of MMP-9RT-PCR:Compared with group A (0.43±0.26),the expression of MMP-9mRNA in group B (0.75±0.44) and group C(0.58±0.32) were significantly increased(P<0.05). Compared with group B,the expression of group C were significantly decreased(P <0.05).6. The results of correlation:There was positive correlation between the protein expression of Galectin-8and MMP-9in airway and lungs.(r=0.89, P<0.05).Conclusions:1、The protein and mRNA expression of Galectin-8and MMP-9in airway and lungs of mice with asthma significantly increased. There was positive correlation between the protein expression of Galectin-8and MMP-9.2、The protein and mRNA expression of Galectin-8and MMP-9in airway and lungs of mice decreased after budesonide treatment.
