| Objective(1)To investigate the cytotoxic effect of Paclitaxel Liposome on multidrug resistance cell line of CNE-1/Taxol;(2)to study the reversal effect of Paclitaxel Liposome on multidrug resistance cell line of CNE-1/Taxol and investigate its possible mechanisms.Methods(1)MTT reduction assay was used to detect the cytotoxic effect of Paclitaxel Liposome on CNE-1/Taxol;(2)MTT reduction assay was used to investigate the reversal effect of Paclitaxel Liposome on multidrug resistance cell line of CNE-1/Taxol, Western blot was used to detect the expression of P-gpã€Bcl-2and Bax.Results(1)MTT method showed that the Paclitaxel Liposome has antitumor activities in vitro on multidrug resistance cell line of CNE-1/Taxol, and it was stronger than Paclitaxel in the same concentration;(2)Paclitaxel Liposome could reverse drug resistance of CNE-1/Taxol cell line to Paclitaxel,and drug resistance index was down regulated from8.43to1.47.It was observed that the expression of P-gp and Bcl-2were down regulates,while Bax was up regulated by using the Paclitaxel Liposome.Conclusion(1)The date suggested that Paclitaxel Liposome possessed stronger antitumor activities than Paclitaxel on CNE-1/Taxol in vitro.(2)Drug resistance of CNE-1/Taxol cell line to Paclitaxel can be reversed by Paclitaxel Liposome.It can be postulated that Paclitaxel Liposome can reverse the drug resistance of CNE-1/Taxol cell line by decrease the expression of P-gp and Bcl-2,increase the expression of Bax. |
