Objective:To investigate the effects of a novel histone deacetylase inhibitor, CC1007on cell cycle and the expression of P21in human cervical carcinoma cells, then discuss the relationship between the block of cell cycle and P21.Methods:1. When Hela, Caski, C33A cells reached the logarithmic growth phase, the cells were exposed to CC1007at different concentrations for24h after which cell cycle analysis was performed with flow cytometer.2. Real-time PCR were undertaken to observe the expression of P21mRNA in Hela, Caski, C33A cells after exposed to CC1007at different concentrations for24h.3. Western blotting were used to detect P21expression in Hela, Caski, C33A cells at logarithmic growth phase after they exposed to CC1007at different concentrations for24h.Results:1. The result of flow cytometer showed that the percentage of the cell population in G1was more increased at higher concentration of CC1007, and the percentage of cells in S was less at lower concentration(P<0.05). The cell populations in G1and S phrase at the concentrations of20μM,30μM,40μM were significantly different from normal group, respectively(P<0.05)..2. We discovered by Real-time PCR that P21mRNA expression were increased with the increase of the concentration, the difference of P21mRNA expression at the concentration of20μM、30μM、40μM being significant (P<0.05).3. The result from western blot showed that expressing of P21protein led to a significant increase with the levels of CC1007concentration, the difference of P21expression at the concentration of20μM、30μM、40μM being significant (P<0.05).Conclusion:1. CC1007can block cells in G1phrase.2. CC1007induce the expression of P21in human cervical carcinoma in a concentration-dependent manner.
