The Expression And Clinical Significance Of S100-A4in Human Glioma

Objective:By analying the expression of S100A4in mRNA and protein of different pathological grade of glioma, so as to provide theoretical basis for the exploration of the mechanism in which way could induce developing glioma. Method:In our experiment, we collected59cases of fresh human glioma specimens,12cases of normal brain tissue specimens and5cases of benign intracranial from Neurosurgery in Xiangya Hospitsl, Central South University in2010-2011. The tissue samples from glioma patients underwent initial surgery and confirmed by pathology, these patients had not received chemotherapy before surgery and had complete related clinical data:the glioma tissue49cases; normal brain tissue12cases; benign intracranial tussue5for example. RT-PCR and Immunohistochemical method(SP) were used to detect mRNA and the protein of the expression of S100A4, the application of the Image J and MIAS medical imageanalysis system software to analysis. SPSS17.0software for statistical analysis. Results:RT-PCR analysis showed that the expression of S100A4mRNA in the normal brain tissue, intracranial benign tumor, and the human brain glioma WHO I,WHO II, WHO III, WHO IV are0.019±0.007,0.0297±0.008,0.1333±0.012,0.1847±0.0133,0.486±0.011,0.5083±0.017.The expression of S100A4in the intracranial benign tumor group and normal group had no difference.(p>0.05). But the expression of S100A4mRNA in the group of normal human brain to glioma grade Ⅰ/Ⅱ, Ⅲ/Ⅳ was gradually increasing (p<0.05). With the increasing of gliomas WHO level,S100A4expression gradually strengthened. Immunohisto-chemical analysis showed that S100A4was expressing mainly in the cytoplasm. The expression rate of S100A4in the normal brain tissue intracranial benign tumor, and the human brain glioma WHO Ⅰ,WHO Ⅱ, WHO Ⅲ, WHO Ⅳ is8.3%,20.0%,33.3%,41.2%,86.4%,90.9%. The expression of S100A4in the intracranial benign tumor group and normal group have no difference. But the expression of S100A4in the group of normal human brain to glioma grade Ⅰ/Ⅱ, Ⅲ/Ⅳ was gradually increasing (p<0.05). With the increasing of gliomas WHO level S100A4expression was gradually strengthening.Conclusion:l.The expression of S100A4is significantly up regulated in primary human brain compared with the intracranial benign tumor and normal brain tissue, S100A4may play promote the development and progression of glioma.2.The expression level of S100A4is significantly positive correlated with the malignant degree of glioma.3.S100A4may become a novel, useful diagnostic and therapeutic marker for glioma.
