Aim:To investigate the relationship of osteoprotegerin (OPG) levels to single nucleotide polymorphism at position950(SNP950) type2diabetes-mellitus(T2DM) and macrovascular complications in Yanbian area in China.Method:we randomly selected230patients in Yanbian area:fifty normal glucose tolerant (NGT) patients and180T2DM patients, which falls into92T2DM patients with carotid artery atherosclerosis and88T2DM patients with carotid artery atherosclerosi. The serum OPG level was obtained using ELISA, and carotid artery IMT was detemtined using Doppler. SNP950of osteoprotegerin gene was genotyped by Taqman-MGB. Their clinical parameters were examined.Results:1. The serum osteoprotegerin level of T2DM was significantly lower than NGT (P<0.01). In the NGT group, the Korean nation was significantly lower than Han nation (P<0.05).2. In age, duration of diabetes, SBP and serum osteoprotegerin level, T2DM with carotid artery atherosclerosis were significantly higher than the T2DM without carotid artery atherosclerosis (P<0.01).3. The frequencies Of CC genotype and C allele in T2DM were higher than those in NGT, but these had no statistically significant (P>0.05).4. T.he frequencies Of CC genotype and C allele in T2DM with carotid artery atherosclerosi were higher than those of T2DM without carotid artery atherosclerosis, but these had not statisticlly significant (P>0.05).5. In NGT group, The frequencies Of CC genotype and C allele in Han nation people were higher than those in korea nation in Yanbian, but these had not statisticlly significant (P>0.05). In T2DM group, these also had not statisticlly significant.6. With logistic regression analysis,the risks of carotid artery atherosclerosis in Yanbian area were: SBP (B=0.050, P=0.043, Wald=4.095, OR=1.051), serum OPG level (B=0.115, P=0.000, Wald=26.592, OR=1.122),and Age (B=0.076, P=0.043, Wald=4.085, OR=1.079)Conclusion:1.The serum osteoprotegerin levels of T2DM with carotid artery atherosclerosis were significantly higher than the T2DM without carotid artery atherosclerosis, serum osteoprotegerin level is an independently significant association with type2diabetes-mellitus macrovascular complications.2. OPG gene SNP950T/C was not associated with T2DM and carotid artery IMT in Yanbian.3. The serum osteoprotegerin levels of T2DM patients were association with age and duration of diabetes, which supports serum osteoprotegerin level may rise with age increased.
