| Chamaecyparis obtusa (Sieb. Et Zucc.) Endl. cv. breviramea f. crippsii, a cultivariety of Chamaecyparis obtusa, is rarely reported as compared with the same-genus plants C. obtusa var. formosana and C. obtusa cv. tetragon, from which a number of natural products with novel structures and biological activities have been isolated. The dissertation consists of two sections. The first chapter is about the studies on chemical and cytotoxic constituents from Chamaecyparis obtuse. The chemical constituents are separated and purificated by means of multiple chromatographic technique (silsca gel, Sephadex LH-20, ODS, HPLC, MCI); their structues are identified by spectra methods of MS, NMR, IR, UV, and so on. Totally,85compounds are obtained from the plant, among which,52compounds have been identified:CD Lignans:Sesamin (1), Savinin (2), Horsfieldin (3),(-)-Dihydrosesamin (4), Dihydrodehydrodiconiferyl alcohol (5), Matairesinol (6),(-)-Arctigenin (7),8-Hydroxypluviatolide (8),8, 8’-Dihydroxyhaplomyrfolin (9), Isolariciresinol-4-O-β-D-Glc (10),(-)-{1R,8S)-4,7,9,3’,9’-Pentahydroxy-3-methoxy-8-4’-oxyneolignan-9’-o-β-D-glucopyranoside (11), Junipercomno sides C(12),(+)-Isolarisiresinol3a-O-β-D-glucopyranoside(13),4,7,9-Trihydroxy-3,3’-dimethoxy-8-O-4’-neolignan-9’-0-β-D-glucopyranoside [7R,8R-threo form](14), Thujastandin-4-O-β-D-glucopyranoside (15),(DTerpenoids: Hinokiol (16),19-Acetoxy-13-hydroxylabada-8(17),14-diene (17),14(R),15-dihydroxylabda acid (18), Libocedrine B (19), Proximadiol (20), Oplodiol monoacetate (21), Oplopanone (22),12-Hydroxyawabukinol (23), Oplodiol (24),4-Eudesmene-1β,11-diol (25),7-Epi-4-Eudesmene-1β,11-diol (26), Chrysanthetriol (27), Corchoionoside C (28),(6S,11R)-14-Hydroxyabscisic acid β-D-glucoside (29),(60)-13-hydroxyabscisic acid0-D-glucoside (30), Megastigm-5-en-3β,9,11-diol (31), Megastigm-5-en-3β,9,11-diol-9-β-D-glucopyranoside (32),(4R)-p-Menth-1-ene-7,8-diol7-O-β-D-glucopyranoside (33),(45)-4-Isopropylcyclohex-1-enecarboxylic acid4-O-β-D-glucopyranoside (34),③Sterols:β-Sitosterol (35), Stigmast-4-en-3-one (36), Daucosterol (37),④Coumarins and Flavonoids:Nordentatin (38), Skimmin(39),7-[[6-O-(6-deoxy-a-Lmannopyranosyl)-β-D-glucopyranosyl]oxy]-2H-1-benzopyran-2-one (40), Quercetin (41), Texifolin (42), Taxifolin-3-O-β-D-glucopyra-noside (43),⑤Others:Dibutylterephthalate (44), Disaccharide (45),1-β-D-glucosyloxy-2-(3-methoxy-4-hydroxyphenyl-propane-1,3-diol)(46),3,4-Dimethoxyphenyl-a-D-glucopyranosid (47), Ferulic Acid-β-D-glucopyranosid (48), Hydrangeifolin I (49),4-(β-D-glucopyranosyloxy) benzoic acid (50),1-Methoxy-6-O-β-apiofuranosylar-butin (51),3-Methoxy O-6-deoxy-a-L-rhamnopyranosyl-(1→6)-β-D-glucopyranoside (52). Thereinto, compounds (9),(15),(23),(30),(31),(32), (34),(51),(52) are new nature products,(1),(2),(3),(4),(16),(17),(18),(35),(37),(40),(41) are found from Chamaecyparis obtusa (Sieb. Et Zucc.) Endl. cv. breviramea f. crippsii and this genus for first time. Selceted compounds1,2,3,4,5,6,16,21,22,25,38,41,42,43were assayed for their anti-cancer activity against human A549and BGC-823cells. Among them, compounds38,41showed anti-cancer activity on A549cells with IC50value of8.079,15.994μg/ml; the compounds2,6,38,41showed anti-cancer activity on BGC-823cells with IC50value of10.755,12.396,6.512,8.974μg/ml; but others showed no anti-cancer activity in the two models。The second chapter is a review on the chemical and bioactive consistuents of Chamaecyparis obtusa. Chamaecyparis obtusa are evergreen trees, more than200compounds have been isolated from them, lots of them are novel compounds and show obviously bioacivities, including antioxidant, anti-cancer and anti-virus activties, and so on. This article presents an overview on main chemical constitents and pharmacological effects of these plants of Chamaecyparis obtusa in domestic and oversea researches, so as to provide a basic scientific evidence for its conservation and sustainable utilization. |
