Study On Extraction And Pharmacological Effects Of Alkali Soluble Polysaccharide In Poria Cocos

Poria cocos is the arid sclerote of poria cocos (Schw.) Wolf which is a kind of mycetes belonging to Polyporaceae. It parasitics and multiplies on Pinaceae pine or pine roots that buried under20-30cm of soil. It is admitted in Chinese Pharmacopeia (2010). Shennong’s herbal regarded it as high grade medicine. Poria cocos can excrete dampness, be used as diuretic to alleviate dropsy, invigorate spleen and calmt he nerves; Modern pharmacology studies have shown that poria hasanti-tumor, regulateimumune,function anti-inflammatory, anti-alergy and other aspects of bio-activity.This thesis belongs to the impact of different concentrations of NaOH, solid-liquid ratio and extraction time on Poria acidic polysaccharide extraction efficiency. The results showed that the optimized extraction of alkali polysaccharideis as follows:the concentration of alkali0.5mol/L, the extracting time1h and the solid-liquid ratio1:70. The main component of Poria cocos is alkali polysaccharide. while very few reports of alkali polysaccharide in Poria cocos.This paper first ivestigated the effect of alkali polysaccharide of Poria cocos on mice immune system. The results show that Prol iferation of lymphocytes from human peripheral blood activated was obviously enhanced by alkali polysaccharide of Poria cocos. Secretions of IL-2、TNF-α and IFN-γ by lymphocytes also were augmented.To further investigate the pharmacological effects of alkali polysaccharide in Poria cocos.This experiment established an optimized rat model of adriamycin-induced nephrotic syngrome, which used two caudal Injection Adriamycin. The results show that alkali polysaccharide in Poria cocos is effective in the treatment of chronic kidney disease in rat induced by adriamycin. Therefore, the results of thisthesis has important significance for interpretation of pharmacological effects of alkal i polysaccharide in Poria cocos. To lay a certain foundation for the depth development of Poria cocos.
