Objective To observe the effect and the possible mechanism of abnormal savdamunziq(ASMq) on morphological structure and serum neurotransmitter levels ofneuroendocrine in depression animal model. Methods Using chronic unpredictable mildstress (CUMs) to establish the depression animal model and four drug groups were givenFluoxetine (3.5mg/kg),three different dosages(1.5、3.0、6.0g/kg.BW) of abnormal savdamunziq respectively during the whole procedure.general stats、changes of morphologicalstructure and the influnce of ASMq to neuroendocrine changes in neurotransmitter levelswhich are Cortisol (CORT)、β-endorphi (β-EP)、Norepinephrine (NE)、Brain-derivedneurotrophicfactor (BDNF)、Adrenocorticotropic hormone (ACTH)、Dopamine(DA)、5-hydroxy tryptamine (5-HT) in the serum of each groups is observed respectively.Results Compared with the normal control group,the depression model group appearedsome general states like slow weight gaining (P<0.05)、 rat hair disorder、lightless,unpeacefulness and low sensitivity to stress; Compared with the depressionmodel group, general states like slow weight gaining(P<0.05)、rat hair disorder、lightless,unpeacefulness and low sensitivity to stress in ASMq three dosage group andFluoxetine were improved significantly.To the morphological structure,model groupanimals show astrocytes edema,endoplasmic reticulum, mitochondrial swelling inHypothalamic; Uneven distribution of pituitary pars cells, reducement of chromophobecells, cell proliferation, secretory granules decreased, nucleolusm marginal,mitochondrial swelling in pituitary; adrenal cortex edema,cell special-shaped,mitochondrial swelling, nuclear shaft widened, cell nuclear matrix fades in Adrenal thanthe normal control group.The morphological changes of ASMq low,middle dosag and theFluoxetine groups like astrocytes edema,endoplasmic reticulum, mitochondrial swellingin Hypothalamic;cell proliferation,nucleolusm marginal,secretory granules decreased inpituitary; adrenal cortex edema,cell special-shaped, mitochondrial swelling in adrenal were improved significantly than the depression model group.the serum level of NE、5-HT、DA、β-EP、BDNF are decreased and the level of ACTH, CORT are increased(P<0.01) in depression model group than the normal control group.Compared with thedepression model group,the serum level of ACTH in ASMq low,middle dosage groupsand the Fluoxetine group were decreased significantly (P<0.05); the serum level ofCORT in ASMq three dosage groups and the Fluoxetine group were decreasedsignificantly (P<0.05), the level of β-EP were increased significantly (P<0.05), the levelof5-HT, DA in ASMq group were increased significantly (P<0.01);the serum level ofBDNF in ASMq middle,high dosage group and the Fluoxetine group were increasedsignificantly (P<0.05); the serum level of NE in ASMq low and the high dosage groupwere increased significantly (P<0.05).Conclusion Abnormal savda munziq can ptetectand recover the morphological damage of hypothalamus、Pituitary and Adrenal in thedepression animal model,the serum level of ACTH and CORT in ASMq group hasdecreased significantly,the serum level of BDNF,β-Ep and Monoamine neurotransmittersin ASMq group has increased significantly,ASMq can also Correct the hyperactivity ofHypothalamus-pituitary-adrenal axis (HPAA) to play antidepressant effect.
