| Objective:The experiment is intended to analyze the relationship between prognosis and vasculogenic mimicry(VM) from follow-up date and prognostic date.Based on these, to investigate what role apoptosis plays in the formation of VM and the potential mechanism.Methods:To collect120prostate cancer resection samples by radical prostatectomy from January11995to December312010of the Second Hospital of Tianjin Medical University. Immunohistochemical staining of CD31(Platelet-endothelial cell adhesion molecule-1) and PAS(Periodic acid-sciff) special histochemical staining were conducted to find VM in prostate cancer.and its morphology and distribution in prostate cancer. To observe the relationship between VM exist and bone metastases according to the follow-up data.Further to observe the different expression of Bcl-2and Caspase-3in VM positive and VM negative tissue, and to explore the effect of apoptosis in the formation of VM.Results:1. From immunohistochemical staining of CD31and PAS special histochemical staining, we observed33cases of VM circuled by tumor cells in the total120cases with the positive rate of27.5%. within which red blood cells can be seen. The boundary membrane of VM is PAS positive, which can be incontinuous or continuous. VM is different from the endothelial vascular,the formation of the VM tumor cell do not secret CD31. There are24cases with bone metastasis in33cases of VM-positive the metastasis rate is72.7%, But there are33cases with bone metastasis in87cases of VM-nagative, the metastasis rate is37.9%, the difference was statistically significant(χ2=11.62, P=0.001).2. In VM-positive group and negative group have shown different levels of Bcl-2and Caspase-3.There are12cases with positive expression of Bcl-2in33cases of VM-positive, the positive rate was36.4%. Whereas there are53cases with positive expression of Bcl-2in87cases of VM-nagative, the positive rate was60.9%. The positive expression rate of Bcl-2in VM-positive group was significantly lower than the VM-negative group, the difference was statistically significant (χ2=5.811, P0.016). There are23cases with positive expression of Caspase-3. in33cases of VM-positive, the positive rate was69.7%. Whereas there are38cases with positive expression of Caspase-3in87cases of VM-nagative, the positive rate was43.7%. The positive expression rate of Caspase-3. in VM-positive group was significantly higher than the VM-negative group, the difference was statistically significant (χ2=6.48P=0.011).Conclusions:1.Vasculogenic mimicry may be a marker of poor prognosis in prostate cancer, bone metastasis are correlated with VM obviously..2.Apoptosisi is related with the formation of VM. The formation of the VM may be dependent on the mitochondrial pathway of apoptosis. |
