Backgroud:Objective: To understand the effect of chicoric acid on the expression of suppressor ofcytokine signaling3(SOCS3) in human heptaocellular carcinoma(HCC) cell line HepG2,the present study investigated the relationships among the mRNA level, proteinexpression of SOCS3and chicoric acid concentration, and the mechanism by whichCRA inhibits the expression of the SOCS3.Methods: human hepatoma cell line was employed for investigation.The cell line ofHepG2was stimulated with a series of concentration of chicoric acid, the SOCS3mRNA was assayed by semi-quantitative RT-PCR and Teal-Time-PCR24h later, andthe expression of SOCS3protein was assayed using Western Blot technique. And then,total protein expression of SOCS3in HepG2treated with different time(0h、6h、12h、18h、24h) of chicoric acid was analyzed using Western Blot. Luciferase reporter assaydetected the SOCS3promoter activity with chicoric acid.Results: Compared with control group,both of mRNA and protein expression ofSOCS3in HepG2cells decreased siganificantly with the increasing concentration ofchicoric acid in a dose-dependent manner (**p<0.01). Moreover, with the increase ofmaniplation time, the protein of expression of SOCS3in HepG2cells was found to bedownregulated significantly by chicoric acid in a dose-and time-dependent way. InHepG2, chicoric acid can inhibits SOCS3promoter activity.Conclusion: The study shows that the chicoric acid can downregulate the expression ofSOCS3in HepG2cells in a dose-and time-dependent way, and the mechanism could beinhibits the promoter activity of SOCS3, this mean chicoric acid inhibits the expressionof SOCS3at transcriptional level. The study on the effect of chicoric acid on SOCS3will provide useful information with regarding to the physiological and pathologicalroles chicroic acid plays in insulin resistance, and suggests a new therapy.
