| Objective: To investigate the effects of different hepatic blood inflow occlusionmaneuverson the growthandmetastasisof livercancer.Methods:Thirty-two Kunming healthy mice were randomly divided into four groups,including SO (sham operation)(n=8), group OPT (occlusion of portal triad)(n=8),groupOPV(occlusion of portal vein)(n=8) and group INT(intermittent clamping)(n=8).Threedays after an hepatocellular carcinoma model was established using portal veininjection,mice underwent either simultaneous clamping of both the portal vein and thehepatic artery or selective clamping of the portal vein to the median and left liver lobesfor60minutes. Sham-operated mice served as controls. Theischemia-reperfusion(I/R),pathological changes of liver tissue,hepatic replacementarea(HRA) and the positive expression rate of the proliferative cell nuclear antigen inhepatocellular carcinoma tissues were measured5days after reperfusion.Results:The OPV group showed significantly lower ALT,AST value in the restorationof blood flow after five days than the OPT group and INT group.Hepatocytes in theOPV group were much less injured than in the OPT group and the INT group inhistopathology.MPO value of SO group was0.2005±0.12(activity unit/g),after24h ofreperfusion, this value for OPT group was0.5304±0.107(activity unit/g),0.386±0.109(activity unit/g)for OPV group and0.5004±0.1(activity unit/g)for INT group(P<0.01).The levels of hepatic replacement area in occlusion of the hepatic lobe in groupsham operation,group occlusion of portal triad, group occlusion of portal vein and groupintermittent clamping were7.658±2.552%,35.612±4.234%,9.02±3.006%and19.946±4.098%respectively(P<0.01). The tumor growth rate of occlusion of thehepatic lobe and nonocclusion of the hepatic lobe was1in group sham operation and2.82in group occlusion of portal triad and1.1in occlusion of portal vein. The positiveexpression rate of the proliferative cell nuclear angiten in hepatocellular carcinomatissues was30%in group SO and78%in group OPT and45%in group OPV and60% in group INT.Conclusion:Presevering hepatic artery flow during portal triad blood inflow occlusionslow down growth and metastasis of liver cancer in mice. |
