| PART I Isolation, Culture and Identification ofBone Marrow-Derived Endothelial Progenitor Cells from RatObjective: To isolate and culture endothelial progenitor cells fromrat’s bone, also explore the conditions of their differentiation intoendothelial cells.Methods: EPCs of SD rat’ s bone were isolated bydensity-gradient centrifugation, then plated on dishes coated withfibronectin for adherent culture, and induced with VEGFã€bFGF and EGF.The attached cells were detected for cell morphology,fluorescence-activated cell sorting, and the cells were also stained with Dil-ac-LDL and FITC-UEA-1.Cellular immunocytochemistry wasperformed to identify the cellsurface markers.Results: Bone marrow sources of EPCs have shape growth, strongability to proliferation. Adherent cells showed that EPCs express CD133,CD34, Flk, â…§ factor (vWF) in different times. The attached cells can stainDil-acLDL and FITC-UEA-1.And we also find w-k body in cytoplasm atthe late stage.Conclusions: Bone can separate marrow and cultivate EPCs by useof a density gradient centrifugation on percoll. EPCs could be differentiatedinto vascular ECs in vitro after induction culture with bFGF, EGF, VEGF,which as the identical feature as the endothelial cells (ECs).
