Objecttive To study the Pharmacokinetics of Cisatracurium in children ofdifferent ages after a single dose.Methods twenty-four ASAⅠorⅡchildren undergoing elective surgery wererandomly divided into2age groups:group A <2yr;group B≥2-<5yr.Atrerial bloodsamples of1ml were obtained at2,4,6,8,10,15,20,30,45,60,90,120min after a bolusdose of Cisatracurium(0.15mg/kg).The plasma concentrations (Cp) of Cisatracuriumwere measured by using high performance liquid chromatograph with fluorescencedetection.The data obtained were analyzed by3P97Pharmacokinetic software.Result The Cp of Cisatracurium decrease rapidly after intravenous bolusinjection in2groups.The curve for Cp versus time was fitted to two-compartmentmodel. There were no significantly differences inT1/2α,T1/2β,A,B,α,β,K10,K12,K21between groups(P>0.05).Group A had a significantlylarger central volume of distribution (Vc),total clearance (CL),apparent volume ofdistribution (Vd) compared with group B(P<0.05).Conclusion The Cp versus time curve for Cisatracurium is best described by atwo-compartment pharmacokinetic model. There were a significantly increase incentral volume of distribution (Vc),total clearance (CL),apparent volume of distribution(Vd) compared with elder children.
