Purpose:To investigate the effect of high volume hemofiltration (HVHF) on high mobility group box chromosomal protein1(HMGB1), TNF-a, IL-6in serum and live,lung, kidney tissues of sepsis dog with acute kidney failure. And to obseve the effect of HVHF on live, lung and kidney tissues.Methods:Twelve male Beagle dogs were ligated of bilateral ureter and injected with lipopolysaccharide(LPS)1.0mg/kg to establish sepsis dog model with acute kidney failure,which were equally randomized to model group and HVHF group(treated with HVHF for24h after injection of LPS immediately).Venous blood was collected to check blood routine,liver and kidney function,electrolyte before and at2,4,8,16,24h after injection of LPS.Arterial blood gas analysis was executed before and at24h after injection of LPS. EILSA was used to determine the concentrations of HMGB1, TNF-a, IL-6in serum at different time points above and in supernatant of live, lung, kidney tissues at24h after injection of LPS. And examine the histologic of lung, live and kidney tissues at24h.Results:General condition:compared with model group, the temperature, the heart and respiratory rate, hypoxia and the bowels are significantly improved in HVHF group. Cytokine levels:compared with model group, the levels of HMGB1, TNF-a, IL-6in serum and live, lung, kidney tissues were decreased (P<0.05), peak time of HMGB1level in serum was delayed.The functions of major organs:compared with model group, the levels of Cr and BUN decline at4h after injected LPS, and the levels of ALT, AST decline at8h (P<0.05). And at24h after injected LPS all dogs obtain respiratory failure, but the PaO2is higher and the PaCO2is lower in HVHF group than the model group (P<0.05)Pathological changes:compared with model group, there is a significant improvement in the histologic of liver and lung tissues (P<0.05); otherwise, the histologic of kidney tissues in two groups no significantly difference (P>0.05)Conclusions:1. HVHF can improve the general condition (the heart and respiratory rate, hypoxia and the bowels) of sepsis dog with acute kidney failure.2. HVHF can decline the concentrations of Cr, BUN, ALT, AST and improve the function of pulmonary of sepsis dog with acute kidney failure.3. HVHF can decrease the concentrations of HMGB1, TNF-a, IL-6in serum and live, lung, kidney tissues and reduce the injury of live, lung, kidney tissues of sepsis dog with acute kidney failure.
