Objective:Through the observation of ffects of electroacupuncture(EA) on morphological structure of important internal organs(the heart, liver and kidney) and the expression of related protein Bcl-2,Bax in rats with Morphine dependence, to explore the mechanism of electroacupuncture therapy to the organic injury of morphine withdrawal.Methods:We selected30SD male rats that weight ranging from260g to300g.Rats were divided randomly into four groups:control groups,model groups,hand acupuncture groups, electroacupuncture groups.The other three groups were injected morphine hydrochloride injection by subcutaneous injection in back day by day (20mg/kg,30mg/kg,40mg/kg,50mg/kg,50mg/kg),five days in all and twice a day (8am,4pm).Three hours after the last injection rats were injected naloxone (3mg/kg) injection by intraperitoneal injection for rapid withdrawal was established by the grade of withdrawal symptoms.We observed different groups rets by different methods: HE staining technique were used to examine morphologic changes Immunohistochemistry technique were used to examine apoptosis of important internal organs and the expression of related protein Bcl-2,Bax and TUNEL method were used to examine cell apoptosis index.Results:1The results of HE staining showed that compared with control group,pathological changes of were more obvious in model group,histomorphological changes of heart,liver,kidney in hand acupuncture group and electroacupuncture group reduced more significantly than those in model group.2The results of TUNEL method shows that compared with control group,the apoptosis index of heart, liver and kidney tissue increase obviously,and there were very significant different (P<0.01);the index in hand acupuncture group and electroacupuncture group reduced significantly than those in model group, and there were very significant different (P<0.01); compared with hand acupuncture group, there were significant different(P 0.05) in those of electroacupuncture group.3The results of the expression of related protein Bcl-2,Bax showed that compared with control group, there were very significant different in model group,hand acupuncture group and electroacupuncture group (P <0.01);in model group,the value of Bax increased obviously and Bcl-2/Bax reduced obviously;in hand acupuncture group and electroacupuncture group, the value of Bcl-2increased obviously and Bcl-2/Bax increased obviously.Compared with model group,there were very significant defferent in hand acupuncture group and electroacupuncture group(P<0.01);compared with hand acupuncture group, there were significant defferent in electroacupuncture group(P<0.05).Conclusions:1Electroacupuncture can promote the expression of Bcl-2and inhibit the expression of Bax, displaying that electroacupuncture can inhibit apoptosis through regulating apoptosis factors.2Electroacupuncture can inhibit the apoptosis of morphine withdrawal, reduce the value of AI, and improve the pathological injure of important organs of bodies.
