Objective To study the expression of CD4~+CD25~+regulatory T cells(Tregs) inperipheral blood and pathological tissue of patients with gastrointestinal malignancy,in order to investigate its value in clinical diagnosis and treatment.Methods The expression of CD4~+CD25~+Fop3~+Tregs and CD4~+CD25~+CD127low/-inperipheral blood was detemined with flow cytometry in60patients withgastrointestinal malignancy (gastric, n=40colonic, n=12rectal, n=8) and20controls.And calculate the ratio of CD4~+CD25~+Fop3~+T/CD4~+T and CD4~+CD25~+CD127low/-/CD4~+T. The expression of CD4~+CD25~+Foxp3~+Tregs in carcinoma tissue and adjacentnoncancerous tissue from the patients was evanuated by immunohistochemicalmethod.Results Comparing with healthy controls, patients with gastrointestinal malignancyin peripheral blood had a higher percentage of CD4~+CD25~+Treg cells. The proportionof CD4~+CD25~+Fop3~+Tregs/CD4~+T in peripheral blood from patients with gastriccancer and colorectal cancer account for (7.46±0.85)%and (7.79±1.19)%respectively,and were both significantly higher than that of healrhy controls(6.65±0.79)%(P<0.01).And the proportion of CD4~+CD25~+CD127low/-/CD4~+T in peripheral blood frompatients with gastric cancer and colorectal cancer account for (7.43±1.19)%and(7.40±1.12)%respectively, and were both significantly higher than that of healrhycontrols(6.22±0.97)%(P<0.01). The average absolute numbers of of Foxp3~+Tregs incarcinoma tissue from patients with gastric cancer and colorectal cancer were(53.52±16.31) and (58.15±14.86) per spuare millimeter, and were both significantlyhigher than that of adjacent noncancerous tissue(6.53±2.68)/mm2(P<0.01). Conclusion The increase percent of regulatory T cells may be tightly related withimmunosuppression and tumor progression in patients with gastrointestinalmalignancy. And high expression of regulatory T cells in histopathological clearancemay be through some kind of mechanism involved in the regulation of tumorimmunity. |
