Objective To explore the expression and clinical significance of the meningioma1(MN1),Homeobox gene5(HOXA5),ETS related gene (ERG) and nucleophosmin(NPM1) in acute myeloid leukemia (AML).Methods MN1, HOXA5, ERG and NPM1expression was measured in216AMLadults by real-time quantitative polymerase chain reaction (RTQ-PCR). It usedstatistical ways to analyze the relationship between MN1, HOXA5, ERG and NPM1expression,also the expressions and age, WBC, HB, PLT, BM blsats, CD34, HLA-DR,CD33. Take looks after the house the gene ABL1to illuminate as the materials forinternal reference.Results(1)Expression level of MN1in threated-AML was lower than de no AML(P<0.05), inM3was lower than non-M3in untreated cases(P<0.05), and the untreated AMLexpression in complete remission was lower than non-remission in one standardinduction therapy(P<0.05). There was no significant difference between the two groupsof60years old-higher with60years old-lower and male with female about the MN1expression in untreated cases(P>0.05).(2)Expression level of HOXA5has no significant difference between threated-AML and de no AML(P>0.05), M3and non-M3(P>0.05), CR-AML and NR-AML(P>0.05).(3)Expression level of ERG has no significant difference between threated-AML and deno AML(P>0.05), M3and non-M3(P>0.05), CR-AML and NR-AML(P>0.05).(4)Expression level of NPM1has no significant difference between threated-AML andde no AML(P>0.05), M3and non-M3(P>0.05), CR-AML and NR-AML(P>0.05).(5)There was no significant correlation in expression level of MN1, HOXA5, ERG andNPM1(P>0.05). Expression level of MN1, HOXA5, ERG and NPM1had nocorrelation with age, hemogram(WBC, HB, PLT), the proportion of bone marrowprogenitor cells, CD34, HLA-DR,CD33.(6)Compete remission rate showed no significant correlation with age, hemogram(WBC, HB, PLT), the proportion of bone marrow progenitor cells, CD34, HLA-DR,CD33, MN1, HOXA5, ERG and NPM1by binary loaistic analyze(P>0.05). MN1,ERG,Age, WBC, PLT,CD34were independent risk factors to outcome in one standardinduction therapy. HOXA5and NPM1were not independent risk factors to outcome inone standard induction therapy, but HOXA5were risk factor together with MN1tooutcome in one standard induction therapy.Conclusion MN1may be an important adverse prognostic factors of AML and animportant indicator in one individualized diagnosis and treatment.HOXA5, ERG, NPM1showed no significant correlation with prognosis of AML.
