Purpose: To evaluation Shengjingsan granule’s clinical curative effect, to observewhether CatSper protein expression or/and CatSper channels function ofAsthenozoospermia patients decrease. Aims to get the scientific and normative clinicaldata, to clarify mechanism of action of Shengjingsan granule, to provide scientific theoryof shengjingsan treatment for Asthenozoospermia, in order to carry forward the essence ofChinese medicine.This experiment furtherly research the role of sperm-specific CatSperchannels in Asthenozoospermia occurred, thus illustrating Asthenozoospermiapathogenesis.It also has an important value to find a new method of clinical treatment orcontraception male.Methods:140patients with asthenospermia were randomly divided into4groups as35cases for Shengjingsan(SJS) treatment group,35cases for placebo treatment group,35cases for Levocarnitine(LCN) treatment group,35cases for control group withouttreatment. Meanwhile30cases normal sperm specimens for normal control group and30cases Asthenozoospermia sperm specimens for Asthenozoospermia group were selected.Human sperm motility was evaluated by computer-assisted semen analysis(CASA) beforeand after a course of treatment were compared between and in the groups. The differencesof the variations in sperm motility of each group before and after the treatment wereinvestigated by statistical method. Wash sperm to prepare for the experiment of Westernblot and Immunohistochemistry.Rusults:(1) Before and after the treatment of Shengjingsan granule, there weresignificant improvements in the rate of rank a(13.05%vs21.37%, P<0.0001),b(19.12%vs25.5%,P<0.01), sperm motility (33.94%vs46.13%,P<0.0001)and sperm viability (43%vs56.15%,P<0.001). There were significant differences between SJS group andplacebo group in the rate of rank a(21.37%vs13.42%,P<0.0001),b(25.5%vs18.38%,P<0.01), sperm motility (46.13%vs31.6%,P<0.0001)and sperm viability(56.15%vs44.48%,P<0.01). There were significant differences between SJS group andcontrol group in the rate of rank a(21.37%vs14.02%,P<0.0001),b(25.5%vs19.64%,P<0.01), sperm motility (46.13%vs33.65%,P<0.0001)and sperm viability(56.15%vs44.47%,P<0.01).(2)Before and after the treatment of LCN, there were significant improvements in therate of rank a(13.68%vs21.6%,P<0.01),b(16.63%vs21%,P<0.001), sperm motility(29.75%vs42.6%,P<0.001)and sperm viabilit(y42.99%vs57.27%,P<0.01). There weresignificant difference between SJS group and placebo group in the rate of rank a(21.6%vs13.42%,P<0.05),b(21%vs18.38%,P<0.01), sperm motility (42.6%vs31.6%,P<0.05)and sperm viability(57.27%vs44.48%,P<0.01). There were significant differencebetween SJS group and control group in the rate of rank a(21.6%vs14.02%,P<0.05),b(21%vs19.64%,P<0.01), sperm motility (42.6%vs33.65%,P<0.05)and spermviability(57.27%vs44.47%,P<0.01).(3) After the treatment of Shengjingsan granule VCL and VSL are significantlybetter than before the treatment (P <0.01). After the treatment of LCN VCL and VSL wassignificantly better than before the treatment (P <0.05). After the treatment ofShengjingsan granule VCL is be superior to placebo treatment group and control groupwithout treatment(P<0.05). After the treatment of Shengjingsan granule VSL is be superiorto placebo treatment group and control group without treatment(P<0.0001). After thetreatment of LCN VCL is be superior to placebo treatment group(P<0.05). After thetreatment of LCN VSL is be superior to placebo treatment group and control group withouttreatment (P<0.0001).(4) normal control group sperm CatSper1protein expression is higher thanAsthenozoospermia group(P<0.01).Conclusions:(1) Shengjingsan granule could improve the rate of rank a,b, spermmotility and sperm viability, so it could improve sperm motility ability to healasthenozoospermia.(2) sperm-specific CatSper channels is possibly relative toasthenozoospermia incidence.
