ObjectiveTo investigate the relationship between insulin levels in follicular fluid and the expression of PTEN in granulosa cells with PCOS (polycystic ovary syndrome)patients, and to provide theoretical basis for understanding the pathogenesis and treatment of PCOS in the future.MethodsA total of51infertile patients who underwent IVF-ET in our centre were recruited in this experiment from Sep2011to Jan2012.31of these patients who suffer from PCOS were selected as experimental group,20cases of non-PCOS patients with tubal obstructions or male factors were selected as control group, meanwhile the experimental group were divided into insulin resistance group (IR group) and non-insulin resistant group (non-IR group). according to homeostasis model assessment of insulin resistance (HOMA-IR). The insulin levels in follicular fluid were measured in all patients, and the expression of PTEN in granulose cells were determined by Immunohistochemical method.Results1) The insulin levels in follicular fluid and the expression of PTEN in granulosa cells was significantly higher in PCOS group than that in control group (P<0.05).2) The BMI、serum testosterone and insulin levels in follicular fluid were significantly higher in IR group compared with those in non-IR group (P<0.05).3) There was a positive correlation between the expression of PTEN in granulosa cells and insulin levels in follicular fluid in IR group (r=0.628, P<0.05), yet there was no significant correlation between the expression of PTEN in granulosa cells and insulin levels in follicular fluid both in non-IR group and non-PCOS group (P>0.05).Conclusion1) The expression of PTEN was significantly higher in IR group than that in non-IR group, and there was a positive correlation between the expression of PTEN in granulosa cells and insulin levels in follicular fluid in IR group, which indicate the high expression of PTEN in granulosa cells may associated with the insulin resistance in ovary and participated in the pathogenesis of PCOS.
