Objective:To evaluate the expression of PHF8in tissue specimens of Laryngocarcinoma and hypopharynx squamous cell carcinoma (LHSCC), and further correlate PHF8expression with clinicopathological parameters and progonisis in LHSCCMethod:PHF8expression in95primary LHSCC tissue specimens were analyzed by immunohistochemistry and correlated with clinicopathological parameters. twelve paired LHSCC tissues were evaluated for PHF8expression by Western-blot.Result:PHF8overexpressed in LHSCC tissues. More importantly, high PHF8expression was significantly associated with tumor site (P=0.001), T classification(P=0.008), clinical stage(P=0.039), recurrence(P=0.023); Kaplan-Meier survival analysis demonstrated that patients with high PHF8expression had both poorer disease-free survival and overall survival than patients with low PHF8expression.Multivariate Cox regression analysis revealed that PHF8overexpression was an independent prognostic factor for patients with LHSCC (P=0.037).Conclusion:This study demonstrated that PHF8protein expression was obviously increased in LHSCC tissue samples, and PHF8protein overexpression was associated with tumor site, recurrence, clinical stage, i ciassilication and poorer prognosis in LHSCC patients. These results suggested that PHF8might play a critical role in the initiation and progression of LHSCC, implicating PHF8as a valuable marker for the prediction of recurrence, and prognosis.
