| ObjectiveOsteosarcoma is a malignant tumor with high degree of malignancy, the prognosis is poor, osteosarcoma developed rapidly, without formal treatment, within one year of lung neoplasm metastasis occurs, resulting in a very low survival rate, after amputation of3~5year survival rate is only5~15%. As with other malignant tumor, osteosarcoma, a serious impact on people health. But osteosarcoma characteristics with other tumor, osteosarcoma is particularly good in a class of people, that is the youth group.More and more long noncoding RNAs (lncRNAs) have been identified recently. These LncRNAs are mRNA-like transcripts, may play an important role in cancer pathomechanisms,but the biological functions of LncRNAs still remain unknown。 In this thesis, we highlight the structure, mechanisms of long non-coding RNA and its functional role in human carcinomas, then we find the most valueable lncRNA to make further research.next we use real time PCR to confirm the resalut,and finaly we analysisthe resault and maybe give a hint to osteosarcoma therapies。Methods This study we used the department established into biological chip company based on8cases of osteosarcoma specimens of the microarray results in Shanghai Kang Cheng corporation, preliminary obtained osteosarcoma of the abnormal expression of genes, based on further analysis of the occurrence and development of in osteosarcoma may play an important role in the gene, thus screens11up-regulated LncRNA expression,12down regulation of LncRNA, the further use of molecular biology experiment real-time PCR verification screening osteosarcoma in the abnormal expression of LncRNA.Result1Using the microarray results of osteosarcoma as the basis, in osteosarcoma tissues we obtained in251abnormal expression genes,68of the genes is up-regulated,183of the genes down-regulated2Combined with the literature analysis, we find out the23genes has the research value, they are: ASLNC21868, ASLNC23518, ASLNC24291, ASLNC23844, exon4713, AW606502, ASLNC21888, DQ786309, BQ183547, BU617029, D20368, ASLNC17568, BF995651, AK026910, ASLNC18376, AK131233, BX649136, ASLNC03769, ASLNC13744, ASLNC19675, ASLNC01877, ASLNC11197, ASLNC21134, BM7206003Using real-time fluorescence quantitative PCR (real-time quantitative PCR) technology to ASLNC01877this gene in osteosarcoma tissues were detected by ASLNC01877, found that the gene expression in osteosarcoma is really in the tissue adjacent to the low, and gene chip in the same direction.Conclusion1using gene chip found in osteosarcoma upregulation or downregulation of the existence of large lncRNA2. Use real-time PCR to a LncRNA verification, the results and the microarray results with the same direction. For the further study of osteosarcoma provides clues. |
