| Objective:To research the effects of17β-estradiol(E2)on secretion of bile acid and expresstion of ATP binding cassette transporter G5(ABCG5) and ATP binding cassette transporter G8(ABCG8) Explore ABCG5and ABCG8estrogen to express the effects of intrahepatic cholestasis of pregnancy in the role.Investigate the effect of E2on the secretion of the bile acid and expression of ATP binding cassette transporter G5(ABCG5) and ATP binding cassette transporter G8(ABCG8) in cultured normal human hepatic cell lines. Explore the role of such an effect that estrogen had on ABCG5/ABCG8expression in intrahepatic cholestasis of pregnancy.Method:Estrogen was added to the cultured normal human liver cell in vitro. The levels of total bile acid in cell culture supernatant which was secreted by liver cell and added to differente concertration E2were measured by ELISA, and the expression of ABCG5/ABCG8of liver cell were examined by Real-Time PCR.Results:1. After control group and cultured normal human liver cell in vitro8h and24h, the level of TBA respectively were (5.92±0.65).(6.25±0.21)umol/L; the expression of ABCG5mRNA respectively were (1.76±0.08)ã€(1.78±0.24); the expression of ABCG8mRNA respectively were (1.73±0.46)ã€(1.79±0.06). The level of TBA and the expression of ABCG5mRNA and ABCG8mRNA,8h compared to24h(P>0.05).2.10nmol/L estrogen and cultured normal human liver cell in vitro8h and24h, the level of TBA respectively were (5.89±0.82)ã€(6.18±0.13) umol/L; the expression of ABCG5mRNA respectively were (1.77±0.56)ã€(1.72±0.3); the expression of ABCG8mRNA respectively were (1.73±0.32)(1.76±0.21). The level of TBA and the expression of ABCG5mRNA and ABCG8mRNA,8h compared to24h(P>0.05).3.0,10,100,500nmol/L estrogen and cultured normal human liver cell in vitro8h,24h, The level of TBA and the expression of ABCG5mRNAand ABCG8mRNA were step down by the estrogen concentration high up (P<0.05). While10nmol/L estrogen compared to control group(P>0.05).100nmol/L estrogen and500nmol/L estrogen compared respectively to control group(P<0.05). prompting that under the action of estrogen, liver cells ABCG5and ABCG8Down-regulated the secretion of bile acid, so they plays an important role on the secretion of bile acid with intrahepatic cholestasis of pregnancy... |
