| Object:Through observations on the effect of Ginseng&Tangerine Peel Concoction (hereinafter called GTPC) on the volume of pepsin, nitric oxide and gastrin level, gastric juice quantity, and ulcer index in acetic acid-induced peptic ulcer rat, the study is to explore the efficacy and action mechanism of GTPC in treating acetic acid induced gastric ulcer, providing experimental bases for the clinical drug application.Method:Acetic acid-induced gastric ulcer rat model was taken into use, and rats were divided at random into6groups, respectively Normal Group, Gastric Ulcer Model Group, Ranitidine group, GTPC High, Medium, and GTPC Low Dose Groups. Observe the effect of GTPC on the volume of pepsin, nitric oxide, gastrin level, gastric juice, and ulcer index in acetic acid-induced peptic ulcer rat. Method of Mett was adopted in determination of pepsin activity; Nitric Acid Reductase was adopted in determination of Nitric Oxide; Radioactive Irmnunoassay was introduced in determination of gastrin level; and Physical Approach was applied in computing ulcer index. Meanwhile, observations were carried out on changes in rats’hair color, weight, food intake and behavior and etc.Result:(1) The establishment of acetic acid-induced gastric ulcer rat model: Scar tissue or ulcerous erosion is seen on the gastric mucosal surface, no presence of gastric perforation.(2) In comparison of the three GTPC Groups(High, Medium, and Low Dose) with Gastric ulcer model group, significant reduction is seen in pepsin activity (P<0.05orP<0.01), bearing distinct difference; In comparison of the GTPC Medium Dose Group with Ranitidine Group, marked reduction is seen in pepsin activity (P<0.05), bearing statistical significance;(3) Nitric Oxide volume in the GTPC High and Medium Dose groups are obviously higher than that of the gastric ulcer model group, which displays conspicuous difference (P<0.05); yet compare GTPC low dose group with gastric ulcer model group, no significant difference is seen in terms of the volume of nitric oxide (P>0.05); while the comparison of the three GTPC Groups (High, Medium and Low Dose) with Ranitidine Group (P>0.05), showed no statistical significance.(4) The volume of Gastrin in the GTPC Groups is significantly lower than that in the gastric ulcer model group (P<0.05), bearing marked difference; in comparison of GTPC High, Low Dose Groups with the gastric ulcer model group, no distinct difference is showed in terms of the volume of Gastrin.(5) The quantity of Gastric Juice in the GTPC Groups is clearly lower than that in the gastric ulcer model group (P<0.05), which indicates marked difference, and GTPC low dose group was found with best efficacy.(6) In comparison of GTPC High, Low Dose Groups with Gastric Ulcer Model Group, distinct reduction in the PH value of gastric juice is seen (P<0.05), manifesting significant difference; The comparison of GTPC High, Low Dose Groups with Ranitidine Group (P<0.05), showed no statistical significance.(7) In comparison of GTPC High, Medium and Low Dose Groups with gastric ulcer model group, significant reduction is seen in terms of ulcer index, which demonstrates apparent difference; while the comparison of those groups with Ranitidine Group (P<0.05), showed no statistical significance.Conclusion:GTPC effectively reduces the gastric mucosal lesions in. acetic acid-induced gastric ulcer rat, significantly bringing down the ulcer index. The mechanism of the GTPC may associate with the approaches to reduce pepsin activity, to increase nitric oxide volume in serum, to inhibit gastrin secretion and to cut down the quantity of gastric juice. Ginseng&Tangerine Peel Concoction shows remarkable effect in the anti-ulcer action. |
