Objective:The protective effect of Niaoduqing particles on the renal lesions of diabetic nephropathy rats and the expression of connective tissue growth factor (CTGF) and transforming growth factor-beta (TGF-β1) on the renal lesions.Methods:29adult male Wistar rats were randomly divided into three groups, normal control group (NC, N=5), diabetic nephropathy in the control group (DN, N=12), Niaoduqing group (N=12). Diabetic nephropathy rat model of production methods: intraperitoneal injection of streptozotocin (STZ,65mg/kg), the establishment of a diabetic nephropathy model. Niaoduqing group was given the Niaoduqing (2.6g/kg/d) gavage normal control group and the DN in the control group given distilled water. In the four weeks of the experiment,8weeks,12weeks to collect24hours urine, to detect the24-hour urinary protein excretion (24UPE) and venous blood in the urine creatinine (Ucr);collecting orbital venous blood, measuring blood glucose (Glu), serum creatinine (Scr) and endogenous creatinine clearance rate calculation (Ccr). All rats were sacrificed in the12th week, the observed changes in the renal histopathology and immunohistochemical method to observe the renal tissue of the connective tissue growth factor (CTGF) and Transforming growth factor-beta (TGF-β1) expression levels.Results:①blood sugar:the DN in the control group, Niaoduqing group rats after12weeks, blood glucose was still significantly higher than the normal control group (P <0.01).②The DN control group compared with normal control group24UPE、Ccr significantly increased (p-<0.05); Niaoduqing group with DN control group comparison, these indicators decreased, with statistical significance (p-<0.05).③In DN group, CTGF and TGF-β1expression was significantly increased in region of glomerular and renal interstitial(p<0.01, compared with NC group); the increases were attenuation in Niaoduqing groups expression (p<0.05, respectively).④The DN group renal pathological changes in the main performance for the kidney small ball size increases, the mesangial matrix, basement membrane thickening; compared with the DN group, the pathological changes of, Niaoduqing group lighter.Conclusions:①Niaoduqing can significantly reduce the expression of CTGF and TGF-β1in renal tissue of diabetic nephropathy rats.②Niaoduqing can delay the progression of diabetic nephropathy.
