Object1To observe dynamically the dominant microbial community from gingival sulcus of normal implants with PCR-DGGE technology;2To learn the differences of the dominant microbial community from gingival sulcus between normal implants and health tooth;3To know the dynamic changes of the dominant microbial community from normal implants gingival sulcus, and their effects on soft tissue around the implants. Then give a reference for the clinical maintenances and health interventions after the dental implant.Methods1Collect the gingival crevicular fluid in the normal implants(Experimental group) and health tooth(Control group) at1month、3months、6months、12months after the dental implant;2Extraction the DNA of gingival crevicular fluid from two groups, to diversity analysis the two groups with PCR-DGGE technology, That is to make electrophoresis and sequencing, and semi-quantitative analysis the measured microorganisms;3Crosswise and vertical comparison the difference of gingival crevicular fluid from the two groups on amount and microbial population.Results1Quantitative-analysis results of gingival crevicular fluidEach sample size is more than0.0040g.Crosswise comparison:each time slot of gingival crevicular fluid from the Experimental group is undifferentiated (P>0.05); Vertical comparison:the gingival crevicular fluid amount from the Experimental group and the Control group are undifferentiated(P>0.05).2The analysis results of PCR-DGGE2.1Crosswise comparison:the fluctuation of diversity index of microbial flora from the Experimental group is greater than the Control group at1-3months, but no difference at6-12months, the diversity index gradually increased as times. Vertical comparison:the fluctuation of diversity index in the Experimental group microbial community is changed, some microbial species disappear or appear at1-3months, and become stabilized at6-12months.2.2The microbial flora of the relative proportion in each period are almost the same both in the Experimental group and the Control group, most of the microbial flora are anaerobe, include porphyromonas gingivalis、eucaryon、 treponema、archaebacteria and so on.2.3The dominant microbial flora in both Experimental group and Control group are almost the same at the constituent ratio, which are streptococcus、 moonpseudoalteromonas、eillonella、evotella shan and collins and so on.Conclusion1The structure of microbial community around dental implant is changed at1-3months, the type and distribution of microbial is variety, and become stabilized at3-12months.2The microbial flora in the normal implants and health tooth are anaerobe primary, but the proportion is low, and its function and pathogenicity need a further research.3The structure changes of microbial community around dental implants are the results of the dominant microbial community self adjusting between the internal species, in order to keep the balance of microbial community around the implants.
