Objective:At present, about half of patients with Parkinson’s disease are from China. The number of Chinese patients with Parkinson’s disease will increase year by year with the aging society. Parkinson’s Disease (PD) is the most common age-related neurodegenerative diseases, which onset after age60the most. Clinically PD patients manifest symptoms of progressive rigidity, bradykinesia, tremor and postural instability. PD is characterized pathologically by the loss of dopaminergic neurons in the substantia nigra pars compacta (SNpc), reduction of dopamine and its metabolite levels and formation of Lewy bodies. The incidence of PD may be associated with genetic and environmental factors, however, the exact pathogenesis is still not clear. The current PD treatment mainly dominated by drug treatment, through the restoration of striatal DA and Ach neurotransmitter system balance in order to relieve symptoms, which can’t block the progression of PD with the appearance of toxic reaction. Acupuncture and moxibustion treatment are widely used in the treatment of PD as a pure natural and safe traditional therapy. In this experiment, to observe behavior changes, the number of TH positive neurons, the activity of mitochondrial complexe Ⅰ-Ⅳ and changes of mitochondrial ultrastructure in PD model rats nigral cell treated by electroacupuncture. And to explore the effects of electroacupuncture treatment on nigral cell mitochondria function and Ultrastructural in rats, and reveal the mechanism of acupuncture treatment on Parkinson’s disease. Methods:SD rats were divided into4groups randomly:a normal group, a solvent control group, a model group and an electroacupuncture therapy group. Model was made by using subcutaneous injection of rotenone in the back of neck.(1)To observe behavior changes of rats, and score the behavior of rats according to the standard of the behavior science.(2) To detect the number of TH positive neurons of all rats by immunohistochemical method.(3)To detect the activity of mitochondrial complex Ⅰ-Ⅳ by ultraviolet spectrophotometry.(4) To observe mitochondrial ultrastructure of rat nigral cell with electron microscope.Result:(1)The abnormol behavior changes of electroacupuncture therapy group is small, while compared with model group (P<0.05).However, normal group and solvent control group have no change.(2) The TH positive neurons count of model group rats is lower than normal group (P<0.05)and solvent control group(P<0.05). Compared with model group, TH positive neuron count of electroacupuncture therapy group is increased significantly (P<0.01)(3) The activity of mitochondrial complex Ⅰ of model group is lower than normal group (P<0.01), and the activity of mitochondrial complex Ⅰ of electroacupuncture therapy group is lower than model group (P <0.01).While the activity of mitochondrial complex Ⅱ,Ⅲ and Ⅳ have no obvious difference.(4) There are no obvious changes in mitochondrial ultrastructure of normal group and solvent control group, and both model group and electroacupuncture therapy group have abnormal change, while the model group rats are serious.Conclusion:Electroacupuncture therapy can can improve the abnormal behaviors and the activity of mitochondrial complexe Ⅰ, and also can block the loss of dopamine neurons in substantia nigra and mitochondrial ultrastructure changes.
