Objective:To study the effects of retinal neurons at hypoxia simulated high altitude,provide a theoretical basis for the retinal damage in the high altitude environment.Methods:Forty-five healthy Wistar rats were randomly divided into650m(the altitude ofUrumchi)altitude group、simulated3000m altitude group and simulated5000m altitudegroup.Rats in simulated3000m altitude group and silulate5000m altitude were put intothe3000m altitude hypobaric chamber and the5000m altitude hypobaric chamberrespectively. All of them underwent ERG, Observed in each group electron microscope,optical microscope, the retina of the morphological changes and determination of thenumber of apoptotic cells at the7th day after bred. Results:1.The amplitudes of ERG bwave and OPs in the simulated5000m altitude group were significantly lower than thosein the650m altitude group. The latencies of ERG and OPs were delayed in the simulated5000m altitude group rats. There were significant differences in the latencies of ERG andOps between the650m altitude group and the silulate5000m altitude group(P<0.05).But there were no significant diffenrences in the latencies of ERG and Ops between thesimulate3000m altitude group and the650m altitude group.2.Observed by lightmicroscope and transmission electron microscope: The inner nuclear layer and retinalnodal cells were arranged disorder in the simulated5000m altitude group rats.No obviouschanges were seen in retinal of the simulate3000m altitude group.3. Observed byTUNEL: The results of TUNEL examination indicated that the massive positive cells werefound in inner nuclear layer and retinal nodal cells in the simulated5000m altitude group.Few apoptosis cells were found in the simulated3000m altitude group, there weresignificant differences between the simulated5000m altitude group and the simulated3000m altitude group(P<0.05).Conclusion:1.There were obvious changes of the rat retinal nerve cell appearance insimulated high altitude hypoxia;2. There were obvious changes of the ERG in simulated high altitude hypoxia.3. At hypoxia simulated high altitude,the apoptosis of retinalneurons was one methods of death in inner nuclear layer and retinal nodal cells.
