Objective: To study the correlation between bacteroides thetaiotaomicron,methanobrevibacter smithii and glucagon-like peptide1(GLP-1) in intestinal in type2diabetes mellitus db/db mice. To explore the possible mechanisms of the effects ofdiabetes. Methods: The type2diabetic mellitus mice group of db/db (n=6) and the controlgroup of db/m (n=6) mice which had been born6weeks were selected to observe theweight and fasting glucose level of mice from the7th to12th week; the12th week laterthen determined the number of bacteroides thetaiotaomicron and methanobrevibactersmithii by RT-qPCR, especially bacteroides thetaiotaomicron by the ways of absolute andrelative quantification with RT-qPCR. The levels of GLP-1on portal blood and colontissue homogenates were measured by enzyme-linked immunosorbent assay (ELISA) toreflect the secretion of GLP-1.The expression of GLP-1R and the level of C-peptide inserum were determined by RT-qPCR and ELISA respectively to reflect the function ofGLP-1. Result: Compared with the control group of db/m,1. the weight of db/db miceand the fasting glucose level increased(P <0.01) from the7th to the12th week. Thechange between glucose and bacteroides thetaiotaomicron in stool was positive;2.thenumber of bacteroides thetaiotaomicron and methanobrevibacter smithii were increasedand two detection means was not statistically significant (P>0.05).3. the levels of GLP-1on portal blood and colon tissue homogenates were reduced (P<0.05).4. The expressionof GLP-1R and the level of C-peptide in serum was down(P<0.05).5. It was negativelycorrelated between the number of bacteroides thetaiotaomicron in colon, and the content of GLP-1on portal blood and in colon(r=-0.63, P<0.01; r=-0.35, P<0.01), and theexpression of GLP-1receptor in pancreas and colon(r=-0.41, P<0.01;r=-0.68, P<0.01); It was negatively correlated between the number of methanobrevibacter smithii incolon, and the content of GLP-1on portal blood and in colon(r=-0.17, P<0.01; r=-0.46, P<0.01), and the expression of GLP-1receptor in pancreas and colon(r=-0.27,P<0.01; r=-0.21, P<0.01). Concludes: The secretion and function of GLP-1inintestinal was related to bacteroides thetaiotaomicron and methanogenic brevibacterium,which prompted that the change in the intestinal flora was concerned with type2diabetes.
