The Fasciola Disease (Fasciolasis) is a zoonotic parasitic disease caused by Fasciola hepatica and Fasciola gigantica infection, was a worldwide distribution. Fasciolosis disease is slow, easy to spread, there has been no effective preventive measures, a serious threat to the pastoral areas of the health of people and livestock development. Therefore, the fasciolosis disease, early diagnosis and immunization has become the hotspot of current research。In this study, bioinformatics analysis of the characteristics of Fasciola gigantica thioredoxin protein peroxidase enzyme (Fg.TPx) gene sequence and its encoded amino acid sequence, primers were designed to amplify the gene Fg.TPx prokaryotic recombinant plasmid, large field Bacillus expression vector for expression and identification。The results showed that the PCR amplification of the length of the target gene is654bp, the gene and the Internet to retrieve the gene sequence homology of98%,99%amino acid sequence homology, expression and purification Fg.TPx molecular weight of about48KDa。The results showed that the Fasciola gigantica natural TPx protein immune activity of recombinant proteins, to maintain a natural epitope of the biological function of TPx, is expected to become a candidate target antigen of Fasciola gigantica disease vaccine, and provides an effective early diagnosis for disease immunodiagnosis of Fasciola gigantica antigenic material。With this recombinant protein as an antigen, the ELISA method, the risk of detection buffaloes infected with Fasciola. Infected with Fasciola positive serum has not been collected, people infected with the of Fasciola disease of the immune diagnostic methods can not carry out, Infected with Fasciola positive serum have not yet gathered, you can not use this method to detect human fasciolosis disease, so we use laboratory-existing monoclonal antibody sandwich ELISA method for detection of circulating antigen, but based on the combination of the two ELISA methods, can be referenced by early diagnosis of infection the Fasciola disease of immunological methods, and his lay the foundation for clinical screening and shaped trematodes infection。...
